Ames tests, in vitro chromosomal aberration tests using mammalian cells, and gene mutation tests gave positive results. In addition, micronucleus tests using fetal mice also produced positive results although it was carried out using unconventional methods (IARC 10, 1976; EHC 80, 1988; NTP DB, accessed in September 2007). Based on these results, the substance was classified into Category 2.

Since the substance is rated as Group 2B by IARC S7 (1987), it was classified into Category 2.

In animal tests, weanling rats were given a single dose of lasiocarpine at 120 mg/kg bw, and up to 18 days after administration, no hepatic lesions were found by autopsy; between 19 and 123 days, delayed hepatic lesions were found by autopsy in 34/49 rats and these 34 rats also showed megalocytosis (EHC 80, 1988). Based on these reports, we concluded that the liver and blood system are target organs of lasiocarpine. The effects of the substance on test animals were seen at doses that fall under the range of Category 1 guidance values. Thus, the substance was classified into Category 1 for the liver and blood system.
