Based on the data which were chosen from the various LD50 values for rats documented in the "List 1"information source designated in the GHS classification guidance for the Japanese government, namely, 2000 mg/kg (male) and 1293 mg/kg bw (female) in the test which was conducted in accordance with OECD test guideline and was reported in the valid information source ("Toxicity Testing reports of Environmental Chemicals" (Chemicals Investigation Promoting Council) (Access on Aug. 2010)), the substance was classified as Category 4.

In addition, about slight extension of estrous cycle, the possibility of the secondary change according to the deterioration of the general status of maternal animals by repeated dose was considered to be the cause. Moreover, the delivery index of 85.85% at 250 mg/kg group was slightly lower than the lower bound (86.74%) of the historical data, and thus, it was guessed to be slight as the influence on reproductive function for maternal animals ("Toxicity Testing Reports of Environmental Chemicals" (Chemicals Investigation Promoting Council), (Access on Aug. 2010)). From the above-mentioned, neither was assumed to be the basis for classification. Therefore, this substance did not induce effects on sexual functions and fertility. In the acute oral toxicity study in rats, deaths occurred at 1000 mg/kg or more, and clinical signs of toxicity such as decrease in locomotor activity, ataxic or abnormal gait, lateral or ventral position, crouching position, lacrimation or chromodacryorrhea, etc. at 1000 mg/kg or higher, and anemia-like symptoms, hypothermia, tremors, hyperirritability and salivation at 2000 mg/kg were observed ("Toxicity Testing Reports of Environmental Chemicals" (Chemicals Investigation Promoting Council), (Access on Aug. 2010)). In addition to these signs, the substance influenced on central nervous system, and it was described that exposure at high levels may result in lowering of consciousness (HSDB (2006)). Thus, the substance was classified as Category 2 (central nervous system). Furthermore, in another acute oral study in rats (LD50 value: 2000 - 2445 mg/kg), clinical signs of lethargy and narcosis were described (PATTY, 5th (2001)). Category 3 (narcotic effects) were, thereby, added. In addition, digestive tract injuries such as erosions and ulcers of the forestomach were reported. However, these findings were excluded from the basis for classification since it was described that those were considered as changes indicating irritating effects on mucous membrane ("Toxicity Testing Reports of Environmental Chemicals" (Chemicals Investigation Promoting Council) (Access on Aug. 2010)).

In the combined repeated oral dose toxicity study with the reproduction/developmental toxicity screening test in rats (OECD TG 422), the principal findings observed at 250 mg/kg/day (converted dose level as that of 90-day study: 97.2 - 117 mg/kg/day) were as follows: decrease in locomotor activity, abnormal gait, suppression of body weight gain in systemic general conditions, low values of erythrocyte count and hemoglobin concentration in hematology, high values of GPT and total bilirubin and low value of triglyceride in clinical chemistry, dark reddish discoloration and enlargement of the spleen, and thickening of forestomach wall in necropsy. Histopathologically, treatment-related changes were observed in the spleen and femoral bone marrow at 250 mg/kg/day, and in the stomach at 50 mg/kg/day or higher. The former included increased erythropoiesis and hemosiderin deposition. The latter consisted of hyperplasia, hyperkeratosis, submucosal edema or inflammatory cellular infiltrations in the squamous epithelium of the forestomach which was attributed to irritability of the test article ("Toxicity Testing Reports of Environmental Chemicals" (Chemicals Investigation Promoting Council), (Access on Aug. 2010)). As stated above, the effects on blood system, spleen and bone marrow appeared, and enhancement of erythropoiesis and increased hemosiderin deposition were noted in the doses equivalent to Category 2 (97.2 - 117 mg/kg/day) of the guidance values ("Toxicity Testing Reports of Environmental Chemicals" (Chemicals Investigation Promoting Council), (Access on Aug. 2010)). Consequently, the substance was classified as Category 2 (hematopoietic system). Besides, the changes in the forestomach observed at 50 mg/kg/day (converted dose level as that of 90-day study: 19.4 - 23.3 mg/kg/day) or more were excluded from the basis for classification because of the judgment as an irritating effect by repeated oral administration with the substance ("Toxicity Testing Reports of Environmental Chemicals" (Chemicals Investigation Promoting Council), (Access on Aug. 2010)).
