In the 2-day inhalation study in rats, the changes reported at the concentration of 104 ppm/4hrs (as vapors: 0.8 mg/L/4hrs) included nose and eye irritation, salivation, dyspnea, swollen lungs in autopsy and lung emphysema in histopathology (IUCLID (2000)). Although the concentration was equivalent to Category 1 judging from the guidance values, the information was derived from the "List 2" information source designated in the GHS classification guidance for the Japanese government and did not meet with the judgment criteria 1b3). Therefore, the substance was classified as Category 2 (respiratory system) in accordance with the guidance document.

In the 14-day inhalation study in rats, which was insufficient for repeated dose toxicity study due to the study using only 4 male rats per group, slight nose irritation, slight thickening of alveolar walls in the lung at 5 ppm (converted dose level as that of 90-day study: 0.0051 mg/L/6hrs), severe nose irritation, dyspnea, thickening of alveolar walls and areas of collapse in the lung at 20 ppm (converted dose level as that of 90-day study: 0.02 mg/L/6hrs) were reported (IUCLID (2000)). Since the dose dependency was confirmed in the multiple parameters with regard to the effects on the respiratory system, the respiratory system was adopted as target organs. Although the concentration was equivalent to Category 1judging from the guidance values, the information was derived from the "List 2" information source designated in the GHS classification guidance for the Japanese government and did not meet the judgment criteria 1b3). Therefore, the substance was classified as Category 2 (respiratory system) according to the guidance document. As relevant information, in the 1-month oral dose study in mice, decline in cholinesterase activity was reported at the dose level of 182 mg/kg/day (converted dose level as that of 90-day study: 60.7 mg/kg/day) (IUCLID (2000)). However, this was not evidenced to classify because no descriptions were given for the other findings such as clinical signs or histological alterations and because the setting of the control group was unspecified.
