Rationale for the classification | |
---|---|---|---|---|---|---|
1 | Acute toxicity (Oral) | Not classified |
- |
- | - | It was classified as "Not classified" in the Classification JIS (corresponding to Category 5 in UN GHS classification) based on an LD50 value of 3,200 mg/kg for rats (IUCLID (2000)). |
1 | Acute toxicity (Dermal) | Classification not possible |
- |
- | - | There is a report on an LD50 value of > 200 mg/kg for rabbits (IUCLID (2000)), but because the category cannot be determined, the classification is not possible. |
1 | Acute toxicity (Inhalation: Gases) | Not applicable |
- |
- | - | Solid (GHS definition) |
1 | Acute toxicity (Inhalation: Vapours) | Classification not possible |
- |
- | - | No data available. |
1 | Acute toxicity (Inhalation: Dusts and mists) | Category 4 |

P304+P340
P261 P271 P312 |
It was classified in Category 4 based on an LC50 value of 1.5 mg/L/4 hours for rats (IUCLID (2000)). Besides, it was regarded as a test on dust because there is a description of exposure to an aerosol, and the LC50 value was above the saturated vapour pressure concentration (0.0017 mg/L). |
2 | Skin corrosion/irritation | Category 1 |
Danger |

It was corrosive in a test by a 4-hour occlusive application to rabbits (IUCLID (2000)), and in a test by a 4-hour occlusive application of an 80% aqueous solution of this substance to rabbits, severe erythema and slight edema remained after eight days, necrosis was also recorded, and it was judged as corrosive (IUCLID (2000)). Therefore, it was classified in Category 1. Besides, it was classified in Xi: R37/38 in EU classification. |
3 | Serious eye damage/eye irritation | Category 1 |
Danger |
H318 |
P305+P351+P338
P280 P310 |
It was assessed as irritating in a test in which this substance was applied to the rabbit eye without dilution (OECD TG 405), but 2/3 of the animals were removed from the test after eight days due to severe irritation, and the remaining animals still showed slight corneal opacity and slight conjunctival redness after 21 days. And it was also assessed as irritating in another test in which this substance was applied to the rabbit eye without dilution, but severe conjunctival redness, vascularization, and staphyloma were still observed after eight days, and there was no full recovery. Therefore, it was classified in Category 1. Besides, it was classified in Xi: R41 in EU classification (EC-JRC (ESIS) (Access on Sep. 2012)). |
4 | Respiratory sensitization | Classification not possible |
- |
- | - | No data available. |
4 | Skin sensitization | Classification not possible |
- |
- | - | It is reported that a positive rate was 70% (7/10), and it was judged as sensitizing in a skin sensitization test with guinea pigs (skin painting test) (IUCLID (2000)), but it was List 2 data, and the test method was not the one approved by OECD. Therefore, it was classified as "Classification not possible." Besides, it was classified in Xi: R43 in EU classification (EC-JRC (ESIS) (Access on Sep. 2012)). |
5 | Germ cell mutagenicity | Classification not possible |
- |
- | - | The classification is not possible due to no in vivo test data. Besides, as for in vitro tests, it was reported to be negative in an Ames test (IUCLID (2000)). |
6 | Carcinogenicity | Classification not possible |
- |
- | - | No data available. |
7 | Reproductive toxicity | Classification not possible |
- |
- | - | In a combined repeated dose toxicity study with the reproduction/developmental toxicity screening test with rats (OECD TG422, GLP), no effects were observed in estrous cycle, copulation index, fertility index, gestation length, and delivery status, but four out of 10 animals died during a gestation period in the high dose group (300 mg/kg), there were a decrease in delivery index and a decreased number of corpus lutea, accompanied by tendencies of low numbers of implantations and total offspring. On observation of lactation, other than total litter loss in one maternal animal found in the 300 mg/kg group, a low number of live offspring due to deaths of maternal animals and low viability index on day 4 due to the total litter loss were observed (Safety Test (Ministry of Economy, Trade and Industry) (Access on Sep. 2012)). The adverse reproductive effects above were not used for the rationale for classification because they were findings only at the high dose where high mortality was seen in maternal animals. Furthermore, because data are insufficient on effects on the development of offspring including teratogenicity, it was classified as "Classification not possible." |
8 | Specific target organ toxicity - Single exposure | Classification not possible |
- |
- | - | No data available. |
9 | Specific target organ toxicity - Repeated exposure | Category 2 (haemal system) |

In a combined repeated dose toxicity study with the reproduction/developmental toxicity screening test with rats (OECD TG422, GLP), 3/12 males and 7/12 females died in the 300 mg/kg group (converted guidance value: about 150 mg/kg/day), and they showed principal symptoms such as salivation, irregular respiration, a decrease in locomotor activity, tonic convulsion, and clonic convulsion. Pathological examination revealed an increase in hemosiderin deposition in the spleen, hemosiderin deposition of Kupffer cells in the liver, and myocardial degeneration in the dead animals and a maternal animal that had a total litter loss in the 300 mg/kg group, and an increase in hemosiderin deposition in the spleen was also observed in males in the 60 mg/kg group (converted guidance value: about 30 mg/kg/day). On hematological examination, decreases in erythrocyte counts, hemoglobin levels, and hematocrit values and increases in platelet counts and reticulocyte counts were observed. From the above, because an increase in hemosiderin deposition in the spleen and hemosiderin deposition of Kupffer cells in the liver at 300 mg/kg/day suggested the possibility of hemolytic anemia, and such a tendency was also observed in males at 60 mg/kg/day (equivalent to the guidance values for Category 2), it was classified in Category 2 (blood system). Besides, signs such as convulsions were only found in the 300 mg / kg group, which exceeded the guidance value for Category 2, and myocardial degeneration seen in the dead animals in the 300 mg/kg group was not observed in the other animals. |

Reliable chronic toxicity data were not obtained. It was classified in Category 1 because it is not rapidly degradable (a degradation rate by BOD: -6, -6, -7% (Biodegradation and Bioconcentration Results of Existing Chemical Substances under the Chemical Substances Control Law, 2008)), and it was classified in Category 1 in acute toxicity. |
