Rationale for the classification
1 Acute toxicity (Oral) Not classified
-
-
- - It was classified as "Not classified" in the Classification JIS (Category 5 in UN GHS classification) based on reports of LD50 values for rats of 2,830 mg/kg (males) and 3,730 mg/kg (females) for the commercial product (purity 93% at minimum) consisting of this substance (> 85%) and its isomer, 2-phenoxypropan-1-ol (CAS RN 4169-04-4) (< 15%) (SIDS (2006), PATTY (6th, 2012)). Besides, the classification was conducted assuming that the toxicity of purities and the isomer less than 15% is comparable to that of this substance.
1 Acute toxicity (Dermal) Not classified
-
-
- - It was classified as "Not classified" based on reports on an LD50 value of > 2,000 mg/kg for rabbits on the commercial product (purity 93% at minimum) consisting of this substance (> 85%) and its isomer, 2-phenoxypropan-1-ol (CAS RN 4169-04-4) (< 15%) (SIDS (2006), PATTY (6th, 2012)). Besides, the classification was conducted assuming that the toxicity of purities and the isomer less than 15% is comparable to that of this substance.
1 Acute toxicity (Inhalation: Gases) Not applicable
-
-
- - Liquid (GHS definition)
1 Acute toxicity (Inhalation: Vapours) Classification not possible
-
-
- - Classification not possible due to lack of data.
1 Acute toxicity (Inhalation: Dusts and mists) Not classified
-
-
- - It was classified as "Not classified" based on reports of an LC50 value (4-hour inhalation of aerosol) for rats of > 5.4 mg/L (OECD TG403) for the commercial product (purity 93% at minimum) consisting of this substance (> 85%) and its isomer, 2-phenoxypropan-1-ol (CAS RN 4169-04-4) (< 15%) (SIDS (2006), PATTY (6th, 2012)). Besides, by assuming that the toxicity of purities and the isomer less than 15% is comparable to that of this substance, because the test concentration is higher than the saturated vapour concentration (0.0426 mg/L), and due to the description of aerosol, a reference value in the unit of mg/L was used for the classification.
2 Skin corrosion/irritation Not classified
-
-
- - It was classified as "Not classified" because no irritation was observed in a skin irritation test with rabbits according to OECD TG 404 (SIDS (2006))
3 Serious eye damage/eye irritation Category 1

Danger
H318 P305+P351+P338
P280
P310
In a Draize test with rabbits (OECD TG 405), it was assessed to be irritating (SIDS (2006)). Corneal opacity, conjunctival redness, and chemosis were observed as findings, and even after 23 days, the animals showed corneal opacity and conjunctival redness and did not fully recover. Therefore, it was classified in Category 1.
4 Respiratory sensitization Classification not possible
-
-
- - Classification not possible due to lack of data.
4 Skin sensitization Not classified
-
-
- - It is reported in SIDS (2006) that it was negative in a Buehler test with guinea pigs (OECD TG 406). Because the test complies with the OECD test guideline as to the number of animals in a test group (20), a method of applying test substance, etc., it was classified as "Not classified."
5 Germ cell mutagenicity Classification not possible
-
-
- - It was classified as "Classification not possible" because it was not possible to classify a substance as "Not classified" according to the revised GHS classification guidance for the Japanese government. As for in vivo, it was assessed to be negative in a micronucleus test with mouse bone marrow cells (SIDS (2006)). As for in vitro, it was negative in a bacterial reverse mutation test and a chromosomal aberration test with peripheral blood lymphocytes of humans (SIDS (2006)).
6 Carcinogenicity Classification not possible
-
-
- - Classification not possible due to lack of data.
7 Reproductive toxicity Classification not possible
-
-
- - Classification not possible due to lack of data. Besides, no effects on fertility were observed in a two-generation reproductive toxicity test with rats in the oral route (drinking water). At the dose where parent toxicity (weight gain reduction, decreases in food consumption and water consumption) was found, decreases in body weight and organ weights and retardation in sexual maturation were observed in offspring, but no effects on fertility after sexual maturation were found (SIDS (2006), PATTY (6th, 2012)). In a developmental toxicity test with rabbits in the oral route (gavage), at the dose where maternal toxicity (weight gain reduction, decreased food consumption, apathy, lateral position, euthanization (1/15 animals)) was seen, skeletal variations but no malformations were observed (SIDS (2006), PATTY (6th, 2012)). Because the above findings are slight changes in developmental parameters, they were not adopted as evidence of the classification.
8 Specific target organ toxicity - Single exposure Category 3 (narcotic effects, respiratory tract irritation)

In gavage administration to rats (SIDS (2006), PATTY (6th, 2012)), staggering, atonia, paresis, loss of pain reflex, and loss of corneal reflex were observed within the range of Category 2 (1,000-2,000 mg/kg), but they were reversible. Therefore, it was classified in Category 3 (narcotic effects). Besides, because respiratory tract irritation and temporary breathing difficulties were observed after exposure to mist (5.4 mg/L) by inhalation in rats (PATTY (6th, 2012), SIDS (2006)), it was classified in Category 3 (respiratory tract irritation).
9 Specific target organ toxicity - Repeated exposure Classification not possible
-
-
- - Classification not possible due to lack of data. Besides, it is described that in 26-week diet administration test with rats, reduced weight gain was observed at the dose (478 mg/kg/day) above the range of Category 2, but toxicity effects were not found within the range for Category 2 (SIDS (2006), PATTY (6th, 2012)), and in a 28-day dermal administration test with rabbits, systemic toxicity effects were not observed at up to the maximum dose (1,000 mg/kg/day: 333 mg/kg/day (converted guidance value)) (SIDS (2006), PATTY (6th, 2012)), Therefore, it corresponds to "Not classified" in the oral and dermal routes. However, because there is no information on the inhalation route, it was classified as "Classification not possible" due to lack of data.
10 Aspiration hazard Classification not possible
-
-
- - Classification not possible due to lack of data.

Rationale for the classification
11 Hazardous to the aquatic environment (Acute) Not classified
-
-
- - From 72-hour ErC50 > 100 mg/L for algae (Desmodesmus subspicatus) (SIDS, 2006), 48-hour LC50 = 370 mg/L for crustacea (Daphnia magna) (SIDS, 2006), and 96-hour LC50 = 280 mg/L for fish (Pimephales promelas) (SIDS, 2006), it was classified as "Not classified."
11 Hazardous to the aquatic environment (Long-term) Not classified
-
-
- - If chronic toxicity data are used, then it is classified as "Not classified" due to rapid degradability (a 28-day degradation rate by BOD = 72% (SIDS, 2006)), and 72-hour NOEC = 12.5 mg/L for algae (Desmodesmus subspicatus) (SIDS, 2006).
If acute toxicity data are used for a trophic level for which chronic toxicity data are not obtained, then it is classified as "Not classified" because it corresponds to "Not classified" in acute toxicity for both crustacea and fish, and it is not water-insoluble (water solubility = 11000 mg/L (SRC, 2013)).
From the above results, it was classified as "Not classified."
12 Hazardous to the ozone layer Classification not possible
-
-
- - This substance is not listed in the Annexes to the Montreal Protocol.
