In a 28-day gavage administration test with rats, at the dose (125 mg/kg/day (converted guidance value: 39 mg/kg/day)) corresponding to Category 2, cell necrosis (males 4/6 animals, females 2/6 animals) was observed in the liver in addition to increased weights and centrilobular hypertrophy of hepatocytes (males 6/6 animals, females 3/6 animals), and marked increases in ALP activity (males and females: a 114% increase (males) and a 140% increase (females) in comparison with the control group) were shown. Changes in the liver were also found at the dose (30 mg/kg/day (converted guidance value: 9.3 mg/kg/day)) corresponding to Category 1, and males showed increased relative weights of the liver, centrilobular hypertrophy of hepatocytes (1/6 animals), and an increase in ALP activity (a 42% increase) (JECDB (Access on October 2013)). However, these effects at the dose were thought to be slight changes at the upper limit of the guidance values for Category 1. From the above, by considering the toxicological significance and frequency of the findings in the liver, it was classified in Category 2 (liver). Besides, in the test, increased relative weights of the kidney and increased eosinophilic body in the tubular epithelium cells were observed only in males at the dose within the guidance value range for Category 2, but these were findings specific to male rats and were thought not to occur in humans. Therefore, the kidney was not adopted as the target organ.
