In a 28-day gavage administration test with rats, effects on the urinary bladder (transitional cell hyperplasia) were observed at the dose (9.3 mg/kg/day (converted guidance value)) corresponding to Category 1, and effects on the liver (increased weights, centrilobular hypertrophy of hepatocytes, higher blood ALT activity), kidney (increased weights, mineralization in the epithelium of the papilla/pelvis, higher blood urea nitrogen), and lung (mineralization in the alveolar septa) were found at the dose (46.7 mg/kg/day (converted guidance value)) corresponding to Category 2 (JECDB (Access on September 2013)). The centrilobular hypertrophy in the liver occurred in all males and females and was accompanied by increased ALT activity (2.4 times higher than in the control group) in males. Furthermore, the authors considered that mineralization in the kidney and lung was not spontaneous changes but those reflecting abnormal mineral metabolism in the blood, and when the similar changes also occur in humans, they may lead to dysfunction of the kidney and lung. From the above, it was classified in Category 1 (urinary bladder), Category 2 (liver, kidney, lung). |
