There are case reports that asthma symptoms were induced by inhalation exposure in humans (SIDS (2006), NICNAS (2001)). Since it was concluded in SIDS (2006) and NICNAS (2001) that this substance was positive in respiratory sensitization, it was classified in Category 1.

There are two sensitization tests with guinea pigs (OECD TG 406, GLP-compliant). In one study, only slight erythema was observed (1/20 animals in the treatment group, 3/20 animals in the control group), therefore, it was judged as non-sensitizing (SIDS (2006)). In the other test, after induction, positive results were obtained for the challenge by intradermal injection, and negative results were obtained for the challenge by epidermal application (SIDS (2006), NICNAS (2001)). In humans, there are both results in patch tests indicating non-sensitizing (SIDS (2006)) and sensitizing (NICNAS (2001)). From these reports, it was determined in SIDS (2006) and NICNAS (2001) that this substance was a sensitizer.
From the above results, although there were both negative and positive results, the positive results were obtained in humans and animals, and it was also concluded that it is sensitizing in assessment reports in other countries. Therefore, it was classified in Category 1.

This substance was irritating to the respiratory tract. There were multiple cases in humans, but no data were available for classification. As for experimental animals, dyspnea, respiratory distress, and nasal discharge by inhalation exposure with rats, and nasal discharge, irregular breathing, sedation, ataxia, dyspnea, diarrhoea, muscular hypotonia and mydriasis by oral administration to rats were reported (SIDS (2006), ACGIH (7th, 2001), NICNAS (2001)). Besides, the findings by oral administration to rats were observed within the concentration range corresponding to Category 2.
Based on these findings, it was considered to be irritating to the respiratory tract by inhalation, and by the oral route, effects on multiple organs were observed, however, effects on specific organs were not confirmed.
From the above, it was classified in Category 2 (systemic toxicity), in addition, Category 3 (respiratory tract irritation).

- - There were no adverse findings by repeated exposure to this substance with humans. As for experimental animals, in a study in which rats were administered by feeding for 90 days, no toxic findings were observed within or lower than the dose range for Category 2 (1,000 ppm: equivalent to 100 mg/kg/day), and even at a dose exceeding Category 2 (3,000 ppm: equivalent to 225 mg/kg/day), only changes in the gastrointestinal tract epithelial lining (necrosis, atrophy) considered to be due to irritation were observed (SIDS (2006), ACGIH (2001), NICNAS (2001)). Therefore, although it was considered to be equivalent to "Not classified" by the oral route, there was no toxic information in other routes, and it was classified as "Classification not possible" due to lack of data. Besides, in the previous classification, it was classified as "Not classified" only by the oral route information.
