As for in vivo, a micronucleus test and a chromosomal aberration test in mouse bone marrow cells after intraperitoneal administration were positive (NTP DB (Access on August)), and a DNA-binding test in the lung, liver, kidney, and stomach after intraperitoneal administration in rats and mice was positive (IARC 106 (2014)). As for in vitro, it was negative from positive and negative results in a bacterial reverse mutation test, a mouse lymphoma test, a chromosomal aberration test, and a sister chromatid exchange test in cultured mammalian cells (IARC 106 (2014), NTP DB (Access on August)). As above, the substance was classified in Category 2 in accordance with the Guidance.

There is no usable human information on the carcinogenicity of this substance.
As for experimental animals, in a 2-year carcinogenicity test using rats or mice in an oral route (gavage), a trend to increase liver tumors (males: sensitivity was reduced due to a lower survival rate, but significant in a trend test), and increased incidence of fibroadenoma in mammary gland (females, a lower dose only) in rats, and increased incidence of liver tumors (hepatocellular adenoma, hepatocellular carcinoma) (males and females) in mice were observed (IARC 106 (2014), NTP TR 237 (1983)). There is information that in rats, sensitivity in statistical analyses (Mantel-Haenszel test, Cochran-Armitage test, Fisher's exact probability test) was reduced due to a lower survival rate, and evidence of carcinogenicity could not be shown. However, it is reported in NTP that increased incidences of liver tumors in mice are clear evidence of carcinogenicity (NTP TR 237 (1983)). On the other hand, IARC considered that there is a significant increased trend also for liver tumors in male rats by a different statistical test (Life table test), and cocluded that from this together with mammary gland tumor in female rats (fibroadenoma, a lower dose only) and liver tumors in male and female mice, there is sufficient evidence of carcinogenicity in experimental animals (IARC vol. 106 (2014)).
As to classification results by other organizations, it was classified in Class C (possible human carcinogen) by US EPA (IRIS Summary (1991)), and in Group 2B by IARC (IARC 106 (2014)).
From the above, the substance was classified in Category 2 for this hazard class.

This substance is irritating to the respiratory tract (HSDB (Access on July 2015)). As for experimental animals, it is written that in oral administration in guinea pigs at 500 mg/kg, morphological changes of liver damage were shown 24 hours to 11 days after administration (IARC 41 (1986)). However, this knowledge was not adopted for the classification due to unknown details.

As for experimental animals, in a 2-week oral administration toxicity test using rats, fatty liver was observed at 300 mg/kg/day (converted to a 90-day equivalent: 33 mg/kg/day) within a range of Category 2 (IARC 41 (1986)).
On top of that, as effects on liver, micro-vacuolization and centrilobular necrosis in the liver were observed at 300 mg/kg/day in a 10-month gavage administration toxicity test using rats, and at 3430 mg/m3 (converted to a Guidance value equivalent: 2.29 mg/L) in a 12-month inhalation toxicity test using rats and 6-month inhalation toxicity test using rabbits (IARC 41 (1986)), all of which were tests on one dose only.
Besides, in a carcinogenicity test using mice in 103-week gavage administration, inflammation, fatty degeneration, necrosis, hepatocytomegaly in the liver were found at the lowest dose of 250 mg/kg/day although above a range of Category 2 (NTP TR 237 (1983)).
As above, effects on the liver were observed in a short-term test, and in longer-term tests, effects on the liver were found at their lowest dose although above a range of Category 2, and the lowest effect level could not be determined.
Therefore, the substance was classified in Category 2 (liver).
