As for in vivo, there are reports that a chromosomal aberration test with rat bone marrow cells in intraperitoneal administration of this substance was positive (DFGOT Vol. 23 (2007)), and that a chromosome examination with peripheral blood lymphocytes of human (patient) dosed with this substance was positive (Initial Risk Assessment Report (NITE, CERI, NEDO, 2008), NTP DB (Access on August 2015)). As for in vitro, it was negative in bacterial reverse mutation tests (Initial Risk Assessment Report (NITE, CERI, NEDO, 2008), NTP DB (Access on August 2015)). Since there is a positive result of in vivo somatic mutagenicity, it was classified in Category 2.

As for experimental animals, in a 13-week toxicity test with rats dosed by drinking water, at 500 ppm (male: 42.2 mg/kg/day, female: 45.7 mg/kg/day) within the range of Category 2, decreased water consumption, decreased body weight gain, reduced relative kidney weight, decreased serum creatinine, and decreased ALP activity were seen, and hematuria and liver cirrhosis in males, and anisonucleosis in the liver, and decreases in serum cholesterol and total protein in females were observed (Initial Risk Assessment Report (NITE, CERI, NEDO, 2008), DFGOT vol. 23 (2007)).
Therefore, it was classified in Category 2 (liver).
Besides, the description on "Antimony and its compounds" in ACGIH which was used as evidence in the previous classification is that pneumonitis, cardiac and hematological diseases were anticipated from acute and chronic exposure to high concentrations (ACGIH (7th, 2001)). Since it was considered to be based on pneumonia caused by antimony fume, pneumoconiosis caused by diantimony trioxide, and heart disease caused by antimony trisulfide, it was not adopted as rationale for the classification.
