It is reported that in a skin irritation test (OECD TG 404) using rabbits, as a result of a semi-occlusive application with an undiluted solution of this substance for 4 hours, it was irritating since well-defined or moderate erythema and slight to moderate erythema were observed (SIDS (2014)). Moreover, it is reported that in another irritation study using rabbits, irritation was observed after 4-hour application (PATTY (6th, 2012)). As for experimental animals, it is reported that in a single oral administration study using rats, moderate sedation, ataxia, mild tremors, and diuresis were observed at 6,830 mg/kg, which is above Category 2 (PATTY (6th, 2012)). Moreover, it is reported that in a single 4-hour inhalation study using rats, labored breathing, exophthalmos and signs of decreased consciousness were observed but no death cases were found (PATTY (6th, 2012)). There is no information on humans. As for experimental animals, 28-day inhalation toxicity tests using rats, mice or dogs were conducted at two concentrations. Since the lower exposure concentration was a vapor and the higher exposure concentration was a saturated vapor, classification was conducted using the guidance value of vapor for the lower concentration and the guidance value of the mist for the higher concentration. In rats, increase in liver weights, mild renal tubular degeneration related to nephropathy inherent in male rats and so on were observed at not less than 26 ppm (0.22 mg/L) (converted guidance value: 0.07 mg/L), which is equivalent to Category 1 based on the guidance value of vapor (Environmental Risk Assessment for Chemical Substances vol.9 (Ministry of the Environment, 2011), SIDS (2014)). In mice, hepatocellular hypertrophy was observed at not less than 25 ppm (0.21 mg/L) (converted guidance value: 0.06 mg/L), which is equivalent to Category 1 based on the guidance value of vapor, and increase in ALT, an absence of, or decrease in, corpora lutea in the ovary and so on were observed at 109 ppm (0.90 mg/L) (converted guidance value: 0.28 mg/L), which is above Category 2 based on the guidance value of mist (Environmental Risk Assessment for Chemical Substances Vol. 9 (Ministry of the Environment, 2011), SIDS (2014)). In dogs, hepatocellular hypertrophy, increase in ALT, increase in alkaline phosphatase and so on were observed at 109 ppm (0.90 mg/L) (converted guidance value: 0.28 mg/L), which is above Category 2 based on the guidance value of mist (Environmental Risk Assessment for Chemical Substances vol.9 (Ministry of the Environment, 2011), SIDS (2014)). Of the above, since the nephrosis is inherent in male rats, it was not adopted as evidence for classification. In addition, although an effect on the liver was observed, no severe effect was observed at the doses equivalent to up to Category 2. Therefore, this substance was classified as "Classification not possible." Besides, in the previous classification, hepatocellular hypertrophy was used as the basis for classification, but it was not determined as a serious effect, therefore, the previous classification result was different from this time. |
