As for experimental animals, 3-month repeated oral dose toxicity tests by drinking water with rats and mice were conducted, and the following is reported: As for rats, an increase in the kidney weight and spleen congestion at or above 10 ppm (male: 1 mg/kg/day, female: 1 mg/kg/day), and an increase in total protein/albumin and the liver weight at or above 30 ppm (male: 3 mg/kg/day, female: 3 mg/kg/day) were observed, and both doses were within the guidance value range for Category 1; decreases in the erythrocyte counts/the hemoglobin amount/the hematocrit value, an increase in the reticulocyte counts, the hepatocyte cytoplasmic vacuolization of the liver, hematopoietic cell proliferation in the spleen, and bone marrow hyperplasia were observed at or above 300 ppm (male: 25 mg/kg/day, female: 27 mg/kg/day), suppression of weight gain, emaciation, clear cell focus of the liver, and a decrease in the epididymal sperm counts were observed at 1000 ppm (male: 65 mg/kg/day, female: 70 mg/kg/day), and these doses were within the range of the guidance value of Category 2; as for mice, an increase in the liver weight was observed at or above 10 ppm (male: 1.5 mg/kg/day, female: 1.2 mg/kg/day) within the guidance value range for Category 1, centrilobular hypertrophy in the liver was seen at or above 300 ppm (male: 41 mg/kg/day, female: 38 mg/kg/day) within the guidance value range for Category 2, and decreases in the erythrocyte counts/the hemoglobin amount/the hematocrit value and an increase in the kidney weight was found at 1000 ppm (male: 110 mg/kg/day, female: 92 mg/kg/day) near the upper limit of the guidance value range for Category 2 (NTP TOX83 (2017)).
From the above, findings of anemia and findings associated with anemia, effects on the liver, and effects on sperm were observed, and it was classified in Category 2 (haemal system, liver, genetic organs (men)).
