[Rationale for the Classification] As for carcinogenicity, there is no available report in humans. Based on the IARC's classification result (1) and test results of (2), (3), it was classified in Category 1B. The category was revised as the result of reviewing the previous classification due to the revision of the classification in 2013 by IARC. [Evidence Data] (1) It is described in IARC that there is sufficient evidence in experimental animals for carcinogenicity. It classified the substance in Group 2A for carcinogenicity by considering mechanism and others (IARC 103 (2013)). As for classification results by other domestic and international organizations, Japan Society for Occupational Health (JSOH) classified it in Group 2A (OEL Documentations (Carcinogenicity classification) (Japan Society For Occupational Health (JSOH) 2016)), and NTP classified it in R (NTP RoC (14th, 2014)). (2) In two tests in mice dermally applied for 99 weeks, increases in skin tumors were observed (IARC 103 (2013)). (3) In an initiation-promotion test in which mice were given a single application on the skin followed by an application of TPA (12-o-tetradecanoylphorbol 13-acetate) as a promoter, an increase in skin tumors was observed (a group applied this substance and TPA: 57% vs vehicle control group and TPA only group: 0%) (IARC 103 (2013)). |
