[Rationale for the Classification]
From data (1), as the result of oral administration in rats, decreases in the numbers of implantations and litters and deceased fertility index were found as reproductive effects, and an increased incidence of various malformations in the external, visceral, and skeletal systems was observed in fetuses as developmental effects. The findings above occurred at the doses where parent animals did not show general toxicity effects, therefore, it was classified in Category 1B for this hazard class. Besides, the category was revised by the use of a new information source.

[Evidence Data]
(1) In a combined one-generation reproductive toxicity study with the developmental toxicity test (OECD TG 414/415, GLP) with rats dosed by gavage, in parent animals, decreases in the numbers of implantations and litters in groups of the low dose (40 mg/kg/day) or higher doses and deceased fertility index in the high-dose group (120 mg/kg/day) were observed as reproductive effects in females. As general toxicity effects in maternal animals, reduced weight gain was observed at the mid dose (80 mg/kg/day) or higher. As developmental effects, an increased incidence of teratogenesis was observed in fetuses of all dosed groups. Therefore, various malformations in the external, visceral, and skeletal system were observed including cleft palate and anasarca in all the dosed groups, head with domed shape and enlarged ventricle in the mid and high-dose groups, and microphthalmia, bent or short tail, kyphosis, bent ribs, agenesis of the pituitary, anencephaly, pelvic dilatation, enlarged ureter, and cryptorchism in the high-dose group (Background Document to ECHA RAC Opinion (2015)).
