[Rationale for the Classification]
From (1), (2), in a two-generation test in rats, at the doses where general toxicity (effects on the live/thyroid) occurred in parent animals, a decrease in survival rate at birth in F1, F2 offspring and dead or urgently slaughtered animals due to dystocia in maternal animals were observed. Moreover, in developmental toxicity tests, at high doses where maternal toxicity was seen, an increased incidence of skeletal malformations in fetuses (3) and growth retardation, delayed sexual maturation, and tooth disorder in liveborn pups (4) were observed. From the above, because this substance caused death or moribund state due to dystocia in maternal animals at the doses without serious general toxicity in maternal animals in two-generation test and one-generation tests in rats, and teratogenicity was observed at the maternal toxicity dose, it was classified in Category 1B for this hazard class. In this re-classification, the category was revised because data were newly obtained from information sources in List 1.

[Evidence Data]
(1) In a two-generation reproductive toxicity test with rats orally dosed (in the diet) (50-600 ppm), in maternal animals of the parent generation, there were three dead animals and one urgently slaughtered animal in a 300 ppm group and three urgently slaughtered animals in a 600 ppm group, all of which were due to dystocia, and it is thought to be due to marked maternal toxicity. In surviving parent animals of the parent and F1 generations, hepatocellular hypertrophy (males and females) and hypertrophy of thyroid follicular epithelium (females) at or above 300 ppm and hypertrophy of thyroid follicular epithelium (males) at 600 ppm were observed. In F1, F2 offspring, a decrease in survival rate at birth was observed at 600 ppm (Risk Assessment Report (Pesticides) (Food Safety Commission of Japan, 2018), JMPR (2006)).
(2) A one-generation test with rats dosed by feeding (25-1,000 ppm) was conducted to confirm reproducibility of dystocia found in the maternal animals in the two-generation test (1). As a result, animals of difficult deliveries were not observed at 300 ppm of the mid dose (animals with dystocia were found in the two-generation test), in a 1,000 ppm group, six females died or were urgently slaughtered, and four of them died at the start of deliveries or within 24 hours after the start of deliveries (Risk Assessment Report (Pesticides) (Food Safety Commission of Japan, 2018), JMPR (2006)).
(3) In developmental toxicity tests with pregnant rats or rabbits dosed by gavage, in rats, at 50 mg/kg/day of the high dose where maternal animals showed transiently reduced weight gain and decreased food consumption, an increased incidence of limb bones dysplasia (humerus, radius, and scapula) was observed in fetuses in addition to increased number of late resorption of embryos, delayed ossification, and skeletal variations (Risk Assessment Report (Pesticides) (Food Safety Commission of Japan, 2018), JMPR (2006)). On the other hand, in a test in rabbits, in a high-dose group of 45 mg/kg/day, abortions (2/24 animals), total resorptions of embryos (3/24 animals) in maternal animals, increased post-implantation loss and delayed ossification in fetuses were observed, but skeletal malformations were not found (Risk Assessment Report (Pesticides) (Food Safety Commission of Japan, 2018), JMPR (2006)).
(4) In a developmental neurotoxicity test in which pregnant rats were orally dosed (by feeding) from gestational day 0 to the weaning of live born (50-500 ppm: 4.4-40.8 mg/kg/day), at or above 300 ppm where reduced weight gain was found in maternal animals, offspring showed reduced weight gain (males and females), delayed preputial separation (males), delayed vaginal patency (females), and at 600 ppm, misalignment of the incisors (males) was observed (Risk Assessment Report (Pesticides) (Food Safety Commission of Japan, 2018), JMPR (2006)).
