P270

P405
P501

P312
[Rationale for the Classification]
Based on (1) and (2), it was irritating to the respiratory tract in humans. Based on (3), effects on the central nervous system and respiratory organs were observed at the dose corresponding to Category 1 in experimental animals. Therefore, it was classified in Category 1 (respiratory organs), Category 3 (narcotic effects). The category was changed from the previous classification by the use of the new information.

[Evidence Data]
(1) As for humans, subjects detected an irritating and objectionable odor at 250 ppm and 500 ppm (ACGIH (7th, 2001)).
(2) There is a description that this substance is irritating to the eyes and mucous membranes (PATTY (6th, 2012)).
(3) In a 6-hour single inhalation exposure test with mice, respiratory disorders, central nervous system symptoms (reduced activity, coordination disorders) were observed by exposure to the vapor of this substance at 2 mg/L (converted 4-hour equivalent value: 2.45 mg/L, corresponding to Category 1) (GESTIS (Access on June 2019)).

[Reference Data, etc.]
(4) This substance dimerizes into dicyclopentadiene (CAS RN 77-73-6) (ACGIH (7th, 2019)). As for dicyclopentadiene, there is a report that in a 4-hour single inhalation exposure test with rats at 1,000 ppm (5.4 mg/L, corresponding to Category 1), irritation of the eyes and nose, dyspnea, muscular incoordination, tremor and hypersensitivity were observed, and all animals died later (DFGOT vol.5 (1993), ACGIH (7th, 2019)). Dicyclopentadiene was classified in Category 1 (central nervous system, respiratory organs), Category 3 (narcotic effects) in the GHS classification in FY 2017.
9 Specific target organ toxicity - Repeated exposure Category 2 (liver, kidney)

Warning
H373 P260
P314
P501
[Rationale for the Classification]
Based on (1), it was classified in Category 2 (liver, kidney).

[Evidence Data]
(1) As a result of a total of 35 exposures of rats to this substance at a mean concentration of 500 ppm (converted guidance value: 0.6 mg/L, within the range of Category 2) for 53 days (7 hours/day), mild centrilobular cloudy swelling of the liver cells and vacuolization of the renal tubular epithelium were observed (PATTY (6th, 2012)).

[Reference Data, etc.]
(2) Since this substance dimerizes into dicyclopentadiene, ACGIH recommended using the TLV for dicyclopentadiene if this substance is concerned (ACGIH (7th, 2019)).
(3) As for dicyclopentadiene, the dimer of this substance, as a result of exposures of rats by inhalation to the vapor for 89 days (7 hours/day, 5 days/week), renal effects were observed at or above 35.2 ppm (converted guidance value: 0.1 mg/L, within the range of Category 1), and chronic pneumonia and bronchiectasis were observed at 73.8 ppm (converted guidance value: 0.2 mg/L, within the range of Category 1). As a result of exposures of mice by inhalation to the vapor for 13 weeks (6 hours/day, 5 days/week), deaths (19/90 animals) were observed at 50 ppm (converted guidance value: 0.1 mg/L, within the range of Category 1), many of them were attributed to pulmonary congestion likely due to the result of pulmonary irritation, and two animals died of renal failure (ACGIH (7th, 2019)).
10 Aspiration hazard *
-
-
- - [Rationale for the Classification]
Classification not possible due to lack of data.
