8 Self-reactive substances and mixtures Type G
-
-
- - There is a chemical group associated with self-reactive properties (P-O) present in the molecule, but it is classified in Class 3, PG II in UNRTDG (UN2329). Self-reactive substances and mixtures, hazards of the highest precedence, were classified as Type G according to the GHS classification guidance for the Japanese government.
9 Pyrophoric liquids *

1 Acute toxicity (Dermal) *
-
-
- - [Rationale for the Classification]
Based on (1), it was classified as "Not classified."
Besides, the category was changed from the previous classification according to the revised GHS classification guidance for the Japanese government.

Liquid (GHS definition). It was classified as "Not classified (Not applicable)."
1 Acute toxicity (Inhalation: Vapours) *
-
-
- - [Rationale for the Classification]
Classification could not be specified based on (1). Therefore, it was classified as "Classification not possible."
Besides, since the LC50 value was lower than 90% of the saturated vapor pressure concentration (about 31,581.4 ppm), the reference value in units of ppm was applied as a vapor with a little mist.

[Evidence Data]
(1) LC50 (4 hours) for rats: > 10,000 ppm (ACGIH (7th, 2001), HSDB (Access on June 2019))

P280
P321
[Rationale for the Classification]
Based on (1), it was classified in Category 2.

[Evidence Data]
(1) This substance exhibits moderate to severe persistent irritation to the skin of rabbits (ACGIH (7th, 2001), PATTY (6th, 2012), HSDB (Access on June 2019)).

5 Germ cell mutagenicity *
-
-
- - [Rationale for the Classification]
Based on (1) and (2), since all of the standard combination tests, including in vivo and in vitro tests, were negative, it was classified as "Not classified."

7 Reproductive toxicity Category 1B

Danger
H360 P308+P313
P201
P202
P280
P405
P501
[Rationale for the Classification]
Based on (1) and (2), no reproductive or developmental effects were observed in the reproduction/developmental toxicity screening test, but in the developmental toxicity test, increased fetal resorption, external abnormalities and skeletal and visceral defects were observed at the dose where no maternal toxicity was observed. Therefore, it was classified in Category 1B. Besides, the classification evidence data were reviewed and the category was changed.

[Evidence Data]
(1) In a reproduction/developmental toxicity screening test (OECD TG 421) with rats administered by gavage, as parental toxicity, thickening of the mucosa of the forestomach and salivation were observed, but no reproductive effects were. As for effects on the offspring, agnathia, low set pinna, microphthalmia and hydronephrosis were observed in 1 dead offspring, but these were considered to be spontaneous malformations and it was determined that there was no effect on the offspring (JECDB (Access on June 2019)).
(2) In a developmental toxicity test with female rats administered by gavage on gestation Days 6-15, gross abnormalities, and skeletal and soft tissue defects were observed at the dose at which maternal toxicity was not observed. In addition, an increased frequency of fetal resorption was observed (PATTY (6th, 2012), Mehlman MA, et al.; Toxicol Appl Pharmacol 72 (1): 11923).
8 Specific target organ toxicity - Single exposure Category 3 (Respiratory tract irritation)

Warning

P261
P271
P312
P405
P501
[Rationale for the Classification]
Based on (1) and (2), it was classified in Category 3 (respiratory tract irritation).

[Evidence Data]
(1) It was described that it was irritating to the eyes, skin and upper respiratory system in humans (PATTY (6th, 2012), HSDB (Access on June 2019)).
(2) In a single inhalation exposure test with rats, profound discomfort (the state of being restless), irritation and respiratory distress were observed (ACGIH (7th, 2001)).
9 Specific target organ toxicity - Repeated exposure Category 2 (eye, nervous system, respiratory organs, bone, hematopoietic system)

Warning
H373 P260
P314
P501
[Rationale for the Classification]
Based on (1) and (2), it was classified in Category 2 (eye, nervous system, respiratory organs, bone, hematopoietic system). Besides, as a result of reviewing the observed effects, the previous classification was changed.

[Evidence Data]
(1) In a 28-day oral administration test with rats, in addition to changes in the forestomach considered to be caused by irritation, hypertrophy of alveolar Type II cells cells was seen at or above 60 mg/kg/day (converted guidance value: 18.7 mg/kg/day, within the range of Category 2), and decreased locomotor activity, tremors, salivation, increases in red blood cell counts, hemoglobin and hematocrit, cataracts, hypertrophy of thyroid follicular epithelium, atrophy of the thymus, alveolar/perivascular infiltration of inflammatory cells and aggregation of foam cells, degeneration of nerve fibers of the sciatic nerve and spinal cord, central chromatolysis of nerve cells in the pons (brainstem) and spinal cord, closure of the epiphyseal plate of bone, decreased extramedullary hematopoiesis in the spleen, and decreased hematopoiesis in the bone marrow, etc. were observed at 250 mg/kg/day (converted guidance value: 77.8 mg/kg/day, within the range of Category 2) (JECDB (Access on June 2019)).
(2) In a 4-week (6 hours/day, 5 days/week) inhalation exposure test with rats, clinical signs of eye irritation and mild reversible striate opacities of the lenses at or above 100 ppm (converted guidance value: 0.11 mg/L, within the range of Category 1), deaths (10%) and mild cataracts at or above 300 ppm (converted guidance value: 0.34 mg/L, within the range of Category 2), deaths (over 70%), and lung inflammation and severe cataracts at 600 ppm (converted guidance value: 0.68 mg/L, within the range of Category 2) were observed (ACGIH (7th, 2001), PATTY (6th, 2012)).

Rationale for the classification
11 Hazardous to the aquatic environment Short term (Acute) Category 3
-
-
H402 P273
P501
This substance quickly hydrolyzes in water to form dimethyl phosphite and methanol. From 48-hour EC50 = 25 mg/L for crustacea (Daphnia magna) for dimethyl phosphite (SIDS, 2004), this substance was classified in Category 3.
11 Hazardous to the aquatic environment Long term (Chronic) Category 3
-
-
H412 P273
P501
This substance quickly hydrolyzes in water to form dimethyl phosphite and methanol. Reliable chronic toxicity data were not obtained for both products.
This substance was classified in Category 3 because it was classified in Category 3 in acute toxicity, and dimethyl phosphite is not rapidly degradable (a degradation rate by BOD: 48% (Biodegradation and Bioconcentration Results of Existing Chemical Substances under the Chemical Substances Control Law, 1997)).
12 Hazardous to the ozone layer Classification not possible
-
-
- - Classification not possible due to lack of data.
