However, information on boric acid (CAS RN 10043-35-3) and borax (Na2B4O7 10 H2O, CAS RN 1303-96-4) is considered to be available as boron compounds. [Evidence Data] (1) In reproductive toxicity tests in which mice were given boric acid by feeding or rats were given boric acid or borax by feeding, infertility due to decreased fertility was observed at doses where no general toxic effects were observed in parental animals. As a result of cross-breeding tests, decreased fertility in males was the cause of infertility in mice, whereas complete infertility was also observed in rats when treated females were mated with control males (Initial Risk Assessment Report (NITE, CERI, NEDO, 2008), ATSDR (2010)). (2) In multiple developmental toxicity tests in which pregnant rats or mice were administered boric acid by feeding during the organogenesis period, an increased incidence of malformations (agenesis or shortening of the 13th rib, dilatation of the lateral ventricles, etc.), increased fetal mortality and neonatal mortality were observed at doses where maternal toxicity (reduced body weight gain, decreased food consumption, etc.) was manifested (Initial Risk Assessment Report (NITE, CERI, NEDO, 2008), ATSDR (2010)). (3) In a test in which pregnant rabbits were administered boric acid by gavage during the organogenesis period, an increase in cardiovascular malformations and increased fetal mortality were reported at a dose where decreased body weight was observed in maternal animals (Initial Risk Assessment Report (NITE, CERI, NEDO, 2008)). |

[Rationale for the Classification] There are no reports on the single exposure to this substance in humans and experimental animals. However, based on (1) and (2), it was classified in Category 1 (respiratory organs). [Evidence Data] (1) Since this substance is rapidly hydrolyzed by contact with water to form hydrochloric acid (CAS RN 7647-01-0), it is highly irritating to the skin, mucous membranes and respiratory tract (ACGIH (7th, 2016)). (2) There are descriptions that chemical pneumonia and pulmonary edema may occur as effects on the respiratory organs by acute inhalation exposure to this substance (HSDB (Access on July 2019)). [Reference Data, etc.] (3) In a study in which rats, mice and guinea pigs were exposed by inhalation at 20, 50 or 100 ppm of this substance for 7 hours/day for 2 days conducted in the 1950s, when the cages were cleaned or changed every 2 hours, all the rats survived at 100 ppm, but all the guinea pigs and 93% of the mice (number of animals not stated) died, and widespread inflammation of the lung and interstitial pneumonia were observed in the dead animals. However, if the cage was not cleaned and changed, all species showed high mortality at all concentrations. It was stated that the cause could be due to animals' licking the substance attached to their skin or paws. It was also described that the feet and mouth of the animals were grossly swollen (ACGIH (7th, 2016)). |
