Because a trend toward an increased tumor incidence was observed in female rats in (1), and the carcinogenicity of this substance was shown, it was classified in Category 2. By taking the results in female rats as limited evidence of carcinogenicity, the classification result was changed.

(1) In carcinogenicity tests by 2-year drinking water administration of this substance to male and female rats and mice, increased incidences of endometrial stromal polyps and endometrial stromal sarcoma were observed in female rats, and the carcinogenicity of this substance was shown in female F344/DuCrj (Fischer) rats. In mice, no increase in tumor incidences due to the administration of this substance was found in males or females, and no carcinogenicity of this substance was shown in male and female Crj: BDF1 mice (Results from Carcinogenicity Studies (Ministry of Health, Labour and Welfare, 1994)).

(2) In a 13-week toxicity test with mice dosed by drinking water, as was the case with rats, a decrease in water intake due to the repellent action and reduced body weight gain were observed at or above 4,000 ppm (800 mg/kg/day, exceeding Category 2), and in a blood biochemical examination, findings indicating nutritional disorders were observed at 8,000 ppm (1,600 mg/kg/day, exceeding Category 2)) (Results from Carcinogenicity Studies (Ministry of Health, Labour and Welfare, 1994)).
(3) In a 104-week toxicity test with rats dosed by drinking water, at 7,200 ppm (900mg/kg/day, exceeding Category 2), eosinophilic changes in the olfactory epithelium of the nasal cavity and retinal atrophy were observed in males and females and bile duct hyperplasia was observed in males (Results from Carcinogenicity Studies (Ministry of Health, Labour and Welfare, 1994)).
(4) In a 104-week toxicity test with mice dosed by drinking water, exfoliation of the epithelium of the renal pelvis was observed in females at 1,000 ppm (200 mg/kg/day, exceeding Category 2), eosinophilic changes in the respiratory epithelium of the nasal cavity were observed at or above 4,000 ppm (800 mg/kg/day, exceeding Category 2), and eosinophilic changes in the olfactory epithelium of the nasal cavity were observed at 16,000 ppm (3,200 mg/kg/day, exceeding Category 2) (Results from Carcinogenicity Studies (Ministry of Health, Labour and Welfare, 1994)).
