[Rationale for the Classification] Based on (1) to (3), tumor incidence was observed in two animal species, but it was not clear evidence in all cases, and therefore, it was classified in Category 2. Based on the new findings, the classification result was changed. [Evidence Data] (1) It was reported that, in a carcinogenicity study with rats dosed by feeding for two years, at 2,500 ppm, a significant increase in the combined incidence of clitoral gland adenoma or carcinoma was observed in females, and a significant increase in the incidence of subcutaneous fibroma or fibrosarcoma (combined) was observed in males, and as for carcinogenicity of this substance, equivocal evidence was obtained in males, and some evidence was obtained in females (NTP TR498 (2002)). (2) In a test with B6C3F1 mice dosed by feeding for 105 or 106 weeks, at 5,000 ppm, a significant increase in the combined incidence of bronchiolar/alveolar adenoma or carcinoma was observed in males, and as for carcinogenicity of this substance, equivocal evidence was obtained in males. Besides, it was reported that, as preneoplastic lesions related to lung tumor observed in males, alveolar epithelial bronchiolization and hyperplasia were observed in males and females (NTP TR498 (2002)). (3) As for the classification results by domestic and international organizations, the DFG classified it in Category 3 (DFG MAK (2007)) based on the data in (1) and (2).
