In an acute oral toxicity test with rats (OECD TG401, GLP), hypoactivity, staggering gait, tremors, clonic convulsions, mydriasis, soiled fur in the peri-genitourinary area, or hypothermia were observed after the next day of administration in the 1,000 mg/kg group, and deaths occurred 3-5 days after administration. In the 2,000 mg/kg group, the animals showed hypoactivity, staggering gait, tremors, or mydriasis on and after the next day of administration, all males died 3-5 days after administration, and all females died 1-3 days after administration. It was classified in Category 2 (central nervous system) because signs such as tremors, clonic convulsions, and mydriasis were found at 1,000-2,000 mg/kg corresponding to the guidance values for Category 2 (JECDB (2003)). In a 28-day repeated oral administration test with rats, in the 50 mg/kg group, increases in ASAT, ALAT, and LDH on blood chemistry examination were observed in males, pathological examination revealed a significant increase in weights and dark red discoloration of the liver, and histological examination showed centrilobular hypertrophy of hepatocytes in males in the 5 and 15 mg/kg groups and males and females in the 50 mg/kg group. And single cell necrosis of hepatocytes in males in the 50 mg/kg group, centrilobular vacuolation in males in the 5 and 50 mg/kg groups, bile thrombi and pericholangitis in males and females in the 50 mg/kg group, and hypertrophy of bile duct cells in males in the 50 mg/kg group were observed (JECDB (2003)). Because liver injuries were seen at or above 5 mg/kg/day (converted guidance value: 1.6 mg/kg/day) corresponding to the guidance values for Category 1 from the above report, it was classified in Category 1 (liver).
