Based on the LC50 (4 hours) value of 4.6 ppm for rats (converted value: 0.022 mg/L) (ACGIH (7th, 2015)), it was classified in Category 1. Since the LC50 value was lower than 90% of the saturated vapor pressure concentration (3,391.89 ppm, converted value: 16.53 mg/L), the reference values in the unit of ppm was applied as vapour without mist. 5 Germ cell mutagenicity Classification not possible
-
-
- - As for in vivo, a micronucleus test and a chromosomal aberration test using the bone marrow of mice were negative (ACGIH (7th, 2015), HSDB(Access on June 2016)). As for in vitro, the results of bacterial reverse mutation tests were positive and negative (ACGIH (7th, 2015). Therefore, it was classified as "Classification not possible" according to the GHS classification guidance for the Japanese government. There are no single exposure data for this substance in humans. In an acute toxicity test using rats, the LC50 value for a single 4-hour inhalation exposure was 4.6 ppm (converted value: 0.022 mg/L), and this value was within the guidance value range for Category 1 (ACGIH (7th, 2015)). Also, it is described that, based on clinical signs and gross pathology, lung was considered to be a target organ (ACGIH (7th, 2015)). Therefore, it was classified in Category 1 (respiratory organs).
Besides, it is reported that in the other study using rats exposed to a saturated vapor pressure concentration of this substance (approximately 1600 ppm) for 3, 10 and 30 minutes, all the rats died due to the single inhalation exposure, and nasal irritation in the observation until the time of death, and the pulmonary damage in the necropsy after death were observed (ACGIH (7th, 2015).

There is no information on humans. In a 2-week inhalation toxicity study in rats (6 hours/day, 5 days/week, 10 times of exposure), the effect on the respiratory organs (Goblet cell hyperplasia in the nasal and paranasal cavity and main bronchi, changes in pulmonary function tests etc.) were reported at 4.1 mg/m3 which is equivalent to Category 1 (converted guidance value: 0.0005 mg/L) or more (DFGOT Vol.17 (2002), ACGIH (7th, 2015)). Therefore, it was classified in Category 1 (respiratory organs). FY2016 GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1))
