5 | Germ cell mutagenicity | Not classified |
- |
- | - | Based on a negative result in an oral rat bone marrow micronucleus test (in vivo somatic cell mutagenicity test) (IARC 71 (1999)), the substance was classified as "Not classified". As relevant information, a negative result is reported in a rat liver UDS test (in vivo genotoxicity tests in somatic cells) (IARC 71 (1999)). From in vitro mutagenicity tests, there are reports of a positive Ames test using Salmonella typhimurium (NTP DB (access on Aug. 2009)), a positive mouse lymphoma test (NTP DB (access on Aug. 2009)) and a positive chromosomal aberration test (NTP DB (access on Aug. 2009)). The substance is an existing chemical substance, for which mutagenicity was established under the provisions of Article 57-3 of the Industrial Safety and Health Law. | FY2009 | GHS Classification Guidance by the Japanese Government (March, 2009) |
6 | Carcinogenicity | Not classified |
- |
- | - | Based on the classification of "Group 3" in IARC (IARC 71 (1999)), the substance was classified as "Not classified". As relevant information, in 2-year feeding tests with rats and mice, there were no effects on survival, but there was a significant positive association between concentration administered and the incidences of a combination of urinary bladder neoplasms for female rats and the high dose to control comparison was also significant for these tumors. No compound-related neoplasms were observed in male rats or mice of either sex. | FY2009 | GHS Classification Guidance by the Japanese Government (March, 2009) |

9 | Specific target organ toxicity - Repeated exposure | Category 2 (kidney) |
Warning |
H373 | P260 P314 P501 |
In a 7-week feeding test with rats, there was an increased in mortality at 500 mg/kg bw (conversion value) and a decrease in body weight gain at 34 and 340 mg/kg bw (conversion value). There were no other significant effects (IARC 29 (1982)). In a 4-month oral test with rabbits given 20 mg/kg bw, there were no significant effects other than changes in clinical biochemical examination such as increase in alkaline phosphatase (IARC 29 (1982)). In 104-week feeding tests with rats and mice (375 and 750 ppm in rats, 750 and 1500 ppm in mice), there was an increase in inflammation in the kidney at 375 ppm (18.75 mg/kg) and epithelial hyperplasia in the kidney at 750 ppm (37.5mg/kg) in male rats with increased incidence of kidney tumors. There was an increase in inflammation in the kidney at 37.5 mg/kg in female rats (IARC 29 (1982)). These levels are within the guidance value range for Category 2. The substance was classified into Category 2 (kidney) since kidney lesions were observed in male and female rats while it is impossible to rule out association with precancerous lesion. | FY2009 | GHS Classification Guidance by the Japanese Government (March, 2009) |
