In a test by an occlusive application of undiluted this substance to the rabbit skin for 3 minutes, 1 hour, or 4 hours, edema, erythema, and necrosis were observed from 1 hour after the application, necrosis that occurred hardened with time and became leather-like, and this persisted until day 8, the end of the observation period. As a result, it was judged as corrosive (SIDS (2003)), therefore it was classified in Category 1C. Besides, it is reported that necrosis in addition to erythema and edema were also found in another test by a 20-hour occlusive application of undiluted this substance to the rabbit skin, and it was assessed to be corrosive (SIDS (2003)). - - In a developmental toxicity test by oral administration to rats on gestational days 6-19 (OECD TG 414, US EPA OPPTS 870.3700, GLP), pre- and postimplantation loss was not changed at any dose, nor was the proportion of the viable fetuses. And fetal external and histopathological examination did not reveal any increase of malformation or variations compared to the control group, and no developmental toxicity or teratogenicity was observed at any dose (50, 150, 300, 450 mg/kg bw/day) (SIDS (2003)). On the other hand, in a one-generation reproductive test by drinking water administration to rats (OECD TG 415, GLP), parent animals did not show systemic toxicity, water consumption and food consumption decreased, but indices for mating, fertility, and litter size and birth and lactation indices were not changed. Therefore, it is reported that there were no effects on reproductive performance, fertility, or the development of offspring at any dose (6, 21, 77 mg/kg bw/day) (SIDS (2003)). From the above reports, no adverse effects on sexual function/fertility or the development of offspring were observed, but because the one-generation reproductive test has a low highest dose and insufficient assessment of the reproductive process, it was questionable to classify as "Not classified." In an acute toxicity test by 4-hour inhalation exposure to concentrations of 0.97-6.08 mg/L (vapour) in rats (LC50: 3.7 mg/L), salivation, nasal discharges, closed lids, labored breathing, and unkempt fur were observed in dosed groups, the signs were more pronounced at higher doses and included dyspnea, unsteady gait, apathia, and tremors (SIDS (2003)). Because the test concentrations were within the guidance value range for Category 1, and edematous lung was described as necropsy findings in dead animals in addition to the signs above, it was classified in Category 1 (respiratory system). Besides, in an acute toxicity test by oral administration to rats (doses: 316, 464, 562, 681, 825, 1,000 mg/kg bw, LD50: 690 mg/kg), symptoms of apathy, dyspnea, salivation, labored breathing, tremors, and unsteady gait were found at doses corresponding to the guidance value range for Category 2 (SIDS (2003)). In a 28-day inhalation exposure test with rats (OECD TG412, GLP), as symptoms of respiratory tract irritation, focal squamous metaplasia of the respiratory epithelium in the nasal cavity and changes in the larynx cartilage at or above 100 ppm/6 hours (converted guidance value: 0.106 mg/L/6 hours), and atrophy of the olfactory epithelium in part of animals at 300 ppm/6 hours (converted guidance value: 0.318 mg/L/6 hours) were observed (SIDS (2003)).
