This substance is irritating to the respiratory tract (ACGIH (7th, 2001), PATTY (6th, 2012), HSDB (Access on September 2014)). There are no human data, however, as for experimental animals, a decreased response to noise at 1,603 ppm (7.49 mg/L), and a decreased respiratory rate and narcosis at 3,207 ppm (14.98 mg/L) by an inhalation exposure with rats (ACGIH (2013), HSDB (Access on September 2014)) were reported. There are no adverse findings by exposure to this substance in humans. However, it is reported that irritation of the mucous membranes, and symptoms in the central nervous system and gastrointestine were observed in occupational exposure to methyl isobutyl ketone (CAS RN: 108-10-1), a substance with a related chemical structure (ACGIH (7th, 2013)).
On the other hand, as for experimental animals, there is a description that in a study in which rats were exposed by inhalation to the vapour of this substance for 2 or 13 weeks, at a concentration of 1,000 ppm (4.66 mg/L: 1 ppm=4.66 mg/m3 (ACGIH (7th, 2013))), nasal and eye irritation, lethargy, decreased aural response, and effects on the kidney (increased weight, regeneration of the tubular epithelium, hyaline droplet formation) were observed (PATTY (6th, 2012), ACGIH (7th, 2013)), and in the 2-week inhalation test, effects on the nasal mucosa, central nervous system and kidney were observed within the range for Category 2 (converted guidance value: 0.62 mg/L/6 hours).
From the above, based on the findings in experimental animals, it was classified in Category 2 (central nervous system, respiratory organs, kidney). Besides, this time, based on the results of the above inhalation exposure tests with rats, which were evidence data for reviewing the TLV value in ACGIH (7th, 2013) after the previous classification, target organs were added. Therefore, the classification result was different from the previous one.
