After occlusive application of 0.5 mL of an undiluted test substance to rabbit ears for 15 or 30 minutes, effects became more serious and the skin was perforated in 1 to 2 days. Surface skin scaling was observed at the end of observation period of 7 days.

Immediately after application of 0.1 mL into a conjunctival sac of rabbits, strong erythema with pronounced chemosis which remained unchanged until the end of the observation period of 7 days.
The evaluation of the cornea, which showed diffuse opacity, was restricted to the first day of the observation period. Afterwards, cornea and iris could not be examined due to edema of the conjunctiva.

Rats dose-dependently showed clinical signs indicating respiratory tract irritation such as bradypnea, dyspnea, labored breathing, and cyanosis by an inhalation exposure at concentrations of 0.007 mg/L/4h (vapour) or higher.
Addition to an occurrence of deaths at 0.056 mg/L/4h (vapour) or higher, significant gross findings in the lungs such as pulmonary edema and hydrothorax observed in necropsy were the only macroscopic detectable effects at the end of the post-observation period (SIDS (2005)).
Also, another inhalation test exposed 0.008 to 0.050 mg/L/4h to rats reported that effects on respiratory tracts were mainly observed, animals died due to respiratory tract lesions at 0.050 mg/L/4h, the prominent microscopic changes such as an increased number of macrophages, perivascular round-cell infiltration, focal fibroproliferative reactions, emphysema, thickening of septa, and abscessive pneumonia were observed at concentrations of 0.025 mg/L/4h or higher, and bradypnea and mucous membrane irritation were observed at 0.008 mg/L/4h (SIDS (2005)).
From the above, it was classified in Category 1 (respiratory system) because a target organ is a respiratory system including respiratory tracts and lungs, and test concentrations were within a value range of Category 1 in Guidance.
Besides, symptoms which are relevant to a respiratory system such as labored breathing and cyanosis were observed at doses of 200 mg/kg or higher in a single dose oral administration test to rats (SIDS (2005)).
