(1) In a reproduction/developmental toxicity screening test (OECD TG 421) with rats administered by gavage, as parental toxicity, thickening of the mucosa of the forestomach and salivation were observed, but no reproductive effects were. As for effects on the offspring, agnathia, low set pinna, microphthalmia and hydronephrosis were observed in 1 dead offspring, but these were considered to be spontaneous malformations and it was determined that there was no effect on the offspring (JECDB (Access on June 2019)).
(2) In a developmental toxicity test with female rats administered by gavage on gestation Days 6-15, gross abnormalities, and skeletal and soft tissue defects were observed at the dose at which maternal toxicity was not observed. In addition, an increased frequency of fetal resorption was observed (PATTY (6th, 2012), Mehlman MA, et al.; Toxicol Appl Pharmacol 72 (1): 11923).

(2) In a 4-week (6 hours/day, 5 days/week) inhalation exposure test with rats, clinical signs of eye irritation and mild reversible striate opacities of the lenses at or above 100 ppm (converted guidance value: 0.11 mg/L, within the range of Category 1), deaths (10%) and mild cataracts at or above 300 ppm (converted guidance value: 0.34 mg/L, within the range of Category 2), deaths (over 70%), and lung inflammation and severe cataracts at 600 ppm (converted guidance value: 0.68 mg/L, within the range of Category 2) were observed (ACGIH (7th, 2001), PATTY (6th, 2012)).
