[Rationale for the Classification] It was classified in Category 1 from (1) based on expert judgment. [Evidence Data] (1) Alkyl (C=12-16) (benzyl) (dimethyl) ammonium chloride (CAS RN: 68424-85-1), a mixture containing this substance, was classified in Category 1 for this hazard class (GHS classification result in FY2020). [Reference Data, etc.] (2) It is reported that as a result of applying 150 microg of this substance to the human skin for 3 days, slight irritation was seen (REACH registration dossier (Accessed Dec. 2020)). |

[Reference Data, etc.] (2) It is reported that in an eye irritation test with rabbits (n = 5), after 15-time applications of a 0.002% solution of this substance at 5-minute intervals, slight conjunctival redness was observed after 4 hours but was fully reversible within 24 hours (REACH registration dossier (Accessed Dec. 2020)). (3) In a test in which eye irritation was evaluated by measuring in-vitro corneal transepithelial electrical resistance (TER) in rabbits, no significant changes in TER were seen in a 0.0025% solution. In the case of 0.005% and 0.01%, TER decreased to 61.1% and 43.7%, respectively, compared with the control group, and this substance exhibited moderate corneal barrier dysfunction (REACH registration dossier (Accessed Dec. 2020)). |

7 | Reproductive toxicity | Classification not possible |
- |
- | - | [Rationale for the Classification] Classification was not possible due to lack of data. [Reference Data, etc.] (1) It was reported that in a two-generation reproduction toxicity study for didecyldimethylammonium chloride (CAS RN 7173-51-5) used as the test substance with rats dosed by feeding, lower body weight and food consumption in parental animals and reduced body weight gain in pups were observed at 100 mg/kg/day. In a subacute oral toxicity test (13 weeks) for benzyltrimethylammonium chloride (CAS RN 56-93-9) used as the test substance with mice dosed by gavage, no effects on male and female reproductive organs, sperm test results, and estrus cycle of females were observed. Based on these findings, the REACH registration dossier indicated that this substance was not likely to be classified as a reproductive toxicant (REACH registration dossier (Accessed Dec. 2020)). |
