---|---|---|---|---|---|---|---|---|
1 | Acute toxicity (Oral) | Category 4 |
Warning |
H302 | P301+P312 P264 P270 P330 P501 |
Three LD50 values are reported for rats (1,300 mg/kg (HSDB (Access on June 2016)), 1,300–4,000 mg/kg (PATTY (6th, 2012)), and 5,770 mg/kg (ACGIH (7th, 2001), PATTY (6th, 2012))). These correspond to Category 4, Category 4–"Not classified" (Category 5 in UN-GHS classification), and "Not classified," respectively. By adopting the category with higher hazard, this substance was classified in Category 4. On the basis of the newly obtained information, the classification was revised. As for the data on IUCLID (2000) and RTECS (2000) which were used for the former classification, these were not adopted because their details are unknown. | FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
1 | Acute toxicity (Dermal) | Not classified |
- |
- | - | On the basis of two reported LD50 values for rabbits of 3,240 mg/kg (ACGIH (7th, 2001), PATTY (6th, 2012)) and 3,970 mg/kg (HSDB (Access on June 2016), this substance was classified as "Not classified" (Category 5 in UN-GHS classification). The classification was revised on the basis of the newly obtained information. | FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
1 | Acute toxicity (Inhalation: Gases) | Not classified (Not applicable) |
- |
- | - | Liquid (GHS definition) | FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
1 | Acute toxicity (Inhalation: Vapours) | Classification not possible |
- |
- | - | As an LC50 value (8 hours) for rats, >2,000 ppm (converted to a 4-hour equivalent value: >2,828 ppm) (ACGIH (7th, 2001)) is reported. However, classification was not possible because a category could not be determined based on this value alone. Besides, a reference value in the unit of ppm was applied as vapor without mist because the LC50 value is lower than 90% of the saturated vapor pressure concentration (3,128 ppm). | FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
1 | Acute toxicity (Inhalation: Dusts and mists) | Classification not possible |
- |
- | - | Classification not possible due to lack of data. | FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
2 | Skin corrosion/irritation | Not classified |
- |
- | - | As topical application of this substance to rabbit skin was associated with only minimal skin irritation (ACGIH (7th, 2001), PATTY (6th, 2012)), it was classified as "Not classified" (Category 3 in UN-GHS classification). | FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
3 | Serious eye damage/eye irritation | Category 1 |
Danger |
H318 | P305+P351+P338 P280 P310 |
Following instillation into the eye of rabbits, this substance caused corneal necrosis (ACGIH (7th, 2001)). Therefore, this substance was classified in Category 1. Although the previous classification was Category 2A on the basis of the same information, the classification was changed to Category 1 based on the description that the substance caused "severe burns with moderately severe corneal necrosis." | FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
4 | Respiratory sensitization | Classification not possible |
- |
- | - | Classification not possible due to lack of data. | FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
4 | Skin sensitization | Classification not possible |
- |
- | - | Classification not possible due to lack of data. Besides, although it is reported that skin sensitization could not be demonstrated in a maximization test on 25 volunteers, the results of this test was not adopted because the details were unknown (GESTIS (Access on June 2016)). | FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
5 | Germ cell mutagenicity | Classification not possible |
- |
- | - | Classification not possible due to lack of data. Besides, the information in IUCLID which was adopted for the former classification could not be verified because it is no longer publicly available. | FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
6 | Carcinogenicity | Classification not possible |
- |
- | - | Classification not possible due to lack of data. Besides, there is a description of a lifetime test in rats which were dosed orally or subcutaneously. The study was inadequate for carcinogenicity assessment (PATTY (6th, 2012)), however. |
FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
7 | Reproductive toxicity | Classification not possible |
- |
- | - | Classification not possible due to lack of data. Besides, there is a description that in a test using pregnant rabbits exposed by inhalation during organogenetic period, no adverse developmental effects were found, even at a high dose level in which maternal toxicity (reduced body weight gain) was observed (PATTY (6th, 2012)). There is nonetheless no information on the effects of this substance on fertility and sexual function. | FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
8 | Specific target organ toxicity - Single exposure | Category 3 (respiratory tract irritation, narcotic effects) |
Warning |
H335 H336 |
P304+P340 P403+P233 P261 P271 P312 P405 P501 |
As for humans, upper respiratory tract irritation is reported in a single inhalation exposure test of this substance on human volunteers (ACGIH (7th, 2001), PATTY (6th, 2012)). In addition, central nervous system depression, weakness, pain, a burning sensation in the chest and stomach, nausea, headache, sleep within 10–15 min, and terminal coma and death within 1h to 6 days are reported in humans who ingested 50–100 mL of this substance (PATTY (6th, 2012)). As for animal experiments, in a single oral exposure study using rabbits, stupor and loss of voluntary movement occurred in one-half of the test animals (ACGIH (7th, 2001)). From the above, this substance was classified in Category 3 (respiratory tract irritation, narcotic effects). | FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
9 | Specific target organ toxicity - Repeated exposure | Classification not possible |
- |
- | - | There is no information on humans. As for experimental animals, in a 3 to 17-week repeated dose toxicity test using rats (the administration route is unknown but is presumably oral administration), in a 1,000 mg/kg/day dose group, there were reported decreases in body weight gain due to reduced food intake and in testes relative weight after 3 weeks, but not after 6 or 17 weeks. The NOAEL is reported as 1,000 mg/kg in this study (HSDB (Access on June 2016)). From the above, the effects that provide the rationale for GHS classification was not seen, even at doses above Category 2 levels. Nonetheless, this substance was classified as "Classification not possible" because hazard-related information was not obtained on other administration routes. |

10 | Aspiration hazard | Classification not possible |
- |
- | - | Classification not possible due to lack of data. Note that the kinematic viscosity of this substance is calculated to be 4.60 mm2/sec (25/15degC) from numerical data (viscosity: 3.74 mPa*s (25degC), density (specific gravity): 0.813 (15/4degC)) listed on HSDB (Access on June 2015). | FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
