The central nervous system was adopted as a target organ from descriptions of a headache, dizziness, prostration, convulsions, coma, and respiratory paralysis as acute poisoning signs caused by oral, inhalation and dermal administration and so on (DHP (13th, 2002)).
And the blood was adopted as a target organ because treatment for methemoglobinemia was referenced as a general treatment for acute poisoning addition to a drop in blood pressure, skin flush, and cyanosis (DHP (13th, 2002)).

From reported neuropsychiatric system disorders such as clonic cramp, stupor, confusion, and motor disorder as serious adverse effects by repeated oral administration of this substance which is a medical antidiarrheal (Ethical Pharmaceuticals (2010), corresponding to List 1) and a description of a possibility of nitrite poisoning (methemoglobinemia, a drop in blood pressure, skin flush) (Ethical Pharmaceuticals (2010)), it was classified in Category 1 (central nervous system, blood).
Besides, findings in the chronic poisoning by repeated administration were mentioned as being similar to the acute poisoning (DHP (13th, 2002)).
