It was judged that based on (1), the symptoms which were considered to be narcotic effects appeared in the oral route, and based on (2), respiratory effects due to respiratory tract mucosa irritation appeared in the inhalation route. Therefore, it was classified as Category 3 (narcotic effects, respiratory tract irritation).

[Evidence Data]
(1) It was reported that in an acute oral toxicity test with rats (OECD TG423, GLP), reduced spontaneous activities (6/6), a decrease of righting reflex (5/6) and piloerection (5/6) were observed from 30 minutes after administration at 300 mg/kg (within the range for Category 1), and all the treated rats died (3/3) at 2,000 mg/kg (within the range for Category 2) and mortalities were preceded by reduced spontaneous activities associated with decreased muscle tone, a decrease or absence of Preyer's and righting reflex, and piloerection (REACH registration dossier (Accessed Aug. 2020)).
(2) It was reported that in an acute inhalation exposure test (dust, exposure to nose) with rats (OECD TG436, GLP, for 4 hours), 1/3 female rats died at 5.32 mg/L (in the range corresponding to "Not classified"). After exposure, all the rats exhibited increased respiratory rate, ataxia, and (in a few instances) noisy respiration which remained one hour post exposure. One day post exposure, all the rats exhibited hunched posture and piloerection. It was reported that the necropsy of the rats that died showed dark patches in the lungs and gaseous distension in the stomach, but the surviving rats showed no abnormalities (REACH registration dossier (Accessed Aug. 2020)).
