[Rationale for the Classification] There were no available reports on carcinogenicity in humans. Based on the existing classification results of (1), and because all results showing carcinogenicity were obtained in two species of experimental animal in (2) and (3), it was classified in Category 1B. [Evidence Data] (1) As for classification results by domestic and international organizations, it was classified in Carc.1B in the EU CLP (EU CLP classification (Access on June 2019)). (2) In a study in which the hydrochloride salt of this substance (CAS RN 6109-97-3) was administered by feeding to rats at 800 ppm or 2,000 ppm for 78 weeks, followed by an observation period, necropsy was performed 107-110 weeks after the start of treatment. Increased incidences of hepatocellular carcinoma in the liver of males and increased combined incidences of hepatocellular carcinoma and neoplastic nodules of the liver of both sexes were observed. In addition, increased combined incidences of squamous cell papilloma, squamous cell carcinoma and basal cell carcinoma in the skin of males, increased combined incidences of carcinoma and squamous cell carcinoma of the Zymbal's gland in the ears of males and females, and increased incidences of adenocarcinoma of the uterus or endometrium, were observed (NTP TR93 (1978)). (3) In a study in which this substance and the hydrochloride salt of this substance were administered by feeding to mice for 78 weeks, increased incidences of hepatocellular carcinomas were observed in male and female treated groups (NTP TR93 (1978)). |
