In the acute oral toxicity study in mice, the LD50 value was 470 mg/kg and abnormal behavior occurred within 2 hours after administration. The changes of clinical signs ranged from general sluggishness at lower dose levels to full anesthesia accompanied by loss of righting reflex and disappearance of the sense of touch at 325 mg/kg or higher. The survivors returned to normal within 24 hours, and the rats treated at 1600 mg/kg or higher persisted coma prior to death (HSDB (2006), the original literature: Leighton FA; Fundam. Appl. Toxicol. 12 (4), 787-792 (1992)). While, it was reported that the substance was potent hepatotoxin and caused serious centrilobular or perilobular necrosis in the liver, which was commonly seen in the substances converted into toxic metabolites in the body (HSDB (2006), the original literature: Leighton FA; Fundam. Appl. Toxicol. 12 (4), 787-792 (1992)). As stated above, the findings after oral administration to mice were observed at the dose levels equivalent to Category 2 of the guidance values.
