5 | Germ cell mutagenicity | Category 2 |
Warning |
H341 | P308+P313 P201 P202 P280 P405 P501 |
From in vivo mutagenicity tests, there are reports of positive results in a micronucleus test and a DNA damage test, however, the details are unknown (RTECS (2008): Mutation Research, vol. 389, p. 3 (1997): Mutation Research, vol. 465, p. 11 (2000)). As relevant information, from in vitro mutagenicity tests, there are reports of positive results in one of two Ames tests, a mouse lymphoma test and a CHO cell chromosomal aberration test (NTP DB (access on Oct. 2009)). Based on these results, the substance was classified into Category 2. | FY2009 | GHS Classification Guidance by the Japanese Government (March, 2009) |
6 | Carcinogenicity | Category 2 |
Warning |
H351 | P308+P313 P201 P202 P280 P405 P501 |
Based on the classification of "Group 2B" in IARC (IARC suppl. 7 (1987)), the substance was classified into Category 2. In one limited epidemiological test, no significant excess of any cancer was observed among 2214 patients who received the substance and were followed for a minimum of 3 years. However, oral administration increased the incidence of hepatocellular adenomas and carcinomas in female mice and induced tumours of the colon and rectum in male and female rats (IARC vol. 24 (1980)). | FY2009 | GHS Classification Guidance by the Japanese Government (March, 2009) |

Danger |
H370 | P308+P311 P260 P264 P270 P321 P405 P501 |
There is a report that toxic signs which occur in humans given large doses of the substance include methaemoglobinaemia, Heinz-body formation, haemolytic anaemia, renal failure, hepatic enlargement, jaundice and hypersensitivity hepatitis (IARC vol. 24 (1980)). Additionally, there are report that methemoglobinemia constituted key feature in 7 human intoxications and that a child ingesting the substance exhibited hepatic enlargement and transiently abnormal renal function (HSDB (2009)). Based on these information, the substance was classified into Category 1 (blood system, liver, kidney). As relevant information, the substance is used as an analgesic drug with local anesthetic activity (IARC vol. 24 (1980)). | FY2009 | GHS Classification Guidance by the Japanese Government (March, 2009) |
9 | Specific target organ toxicity - Repeated exposure | Category 2 (kidney) |
Warning |
H373 | P260 P314 P501 |
Based on reports that calculi composed of the free compound were found in the urinary tract of a 67 year old man who had been taking the substance daily for 2 years, and that the free compound was deposited on ureteral stones already present in a 55 year old man with chronic prostatitis who had taken the substance three times a day for 5 months (HSDB (2009)), the substance was classified into Category 2 (kidney). As relevant information, in a 45-day oral test in rats and mice, there was no gross pathological abnormalities (NTP TR99 (1978)). | FY2009 | GHS Classification Guidance by the Japanese Government (March, 2009) |
