In a 28-day gavage administration test with rats, increased liver weights (males), centrilobular hypertrophy of hepatocytes (males 4/6 animals), and pigmentation in the tubular cells (males 1/6 animals) in the kidney were observed at the dose (20 mg/kg/day (converted guidance value: 6.2 mg/kg/day)) corresponding to Category 1, and at the dose (100 mg/kg/day (converted guidance value: 31.1 mg/kg/day)) corresponding to Category 2, the above pathological findings in the liver and kidney were found in all males and females, and increased weights of the liver and kidney were seen in both males and females (JECDB (Access on September 2011)). The effects on the liver and kidney did not show clear organ toxicity and were normally rejected because they were not serious effects. However, these were adopted this time because there was a clear dose-response relationship in increased weights and histological changes, and they were observed in all males and females in the high dose group. Therefore, it was judged as appropriate to classify it in Category 2 (liver, kidney). Furthermore, in the high dose group, effects on the testis (decreased intraductal spermatid and luminal cell debris in the epididymis, retained spermatid, degeneration/necrosis of the germ cells, a decreased number of pachytene spermatocytes in the seminiferous tubules, etc. in the testis) were also found (JECDB (Access on September 2011)).
