- - There was an oral test in which the substance was administered to rats via drinking water from seven weeks before mating to the gestation period (the date of end of administration was unknown), and fetuses were examined at the day of 12 and 22 of gestation and after delivery. There were no overt signs of toxicity in the treated maternal rats. As for fetuses, only slight effects such as reduction of crown-rump length and somite number, and delayed ossification were observed. Neither the number of litter, nor body weight was different from the control group, and no serious effects on the exterior and a visceral malformation were observed (EHC 200 (1998)). However, there was no reference about the exposure to sires, and no information was provided regarding sexual functions and fertility of parental animals. No data were available for this substance, but the following was information on soluble copper compounds. According to the case reports of ingestion of a large amount of compounds by suicide attempt or accident, clinical signs observed included epigastric pain, headache, nausea, dizziness, vomiting, diarrhea, tachycardia, tachypnea and dyspnea were observed (EHC 200 (1998), PIM G002 (1999)). And it was reported that liver damage (jaundice, elevation of serum transaminase and serum bilirubin levels, enlargement, centrilobular necrosis and cholestasis), renal dysfunction (elevated blood urea nitrogen and urobilinogen levels, proteinuria, anuria and oliguria), and methaemoglobinemia, hemolytic anemia and cyanosis were observed (PIM G002 (1999)). Based on these findings, the substance was classified as Category 1 (liver, kidney, blood). Furthermore, it was described that exposure to copper fumes cause sneezing, coughing and discomfort of gastrointestinal tract (PIM G002 (1999)), and that inhalation of dust cause irritation of throat and lung (HSDB (2009)). In the 105-day feeding study in rabbits, the effects on the liver (pigment deposition, necrosis and cirrhosis) were reported at dietary levels between 97 to 2370 ppm (dose levels: approx. 2.9 - 71 mg/kg/day) (JECFA 17 (1982)). Moreover, as the effect on humans, there were case reports of liver failure following chronic ingestion of drinking-water containing high copper concentrations (EHC 200 (1998)), and a case of jaundice and hepatomegaly following 3-year oral intake of copper supplement (ATSDR (2004)).
