[Rationale for the Classification] Based on (1) and (2), it was classified in Category 1. [Evidence Data] (1) Following contact with the skin, burning sensation, reddening, severe irritation, and painful tissue damage may occur. The extent of these symptoms is assumed to be dependent on the concentration and duration of exposure (GESTIS (Access on September 2019)). (2) This substance is irritating (HSDB (Access on September 2019)). |

[Rationale for the Classification] Based on (1)-(3), it was classified in Category 1. [Evidence Data] (1) This substance is irritating (HSDB (Access on September 2019)). (2) Following eye contact with the pure grade substance or concentrated solutions, tissue damage and corneal opacity as well as a serious burning sensation and blurred vision should be expected as a consequence of the corrosive action (GESTIS (Access on September 2019)). (3) This substance was classified in Category 1 (skin corrosion). |

- | - | [Rationale for the Classification] Classification not possible due to lack of data. [Reference Data, etc.] (1) It was reported to cause morphological changes in the testes when rats were given sodium fluoroacetate (CAS RN 62-74-8) by drinking water for 21 days (ACGIH (7th, 2001)). |

8 | Specific target organ toxicity - Single exposure | Category 1 (central nervous system, cardiovascular system, respiratory organs) |
Danger |
H370 | P308+P311 P260 P264 P270 P321 P405 P501 |
[Rationale for the Classification] Based on (1) and (2), it was classified in Category 1 (central nervous system, cardiovascular system, respiratory organs). Although the kidney was also adopted as a target organ in the previous classification, it was not adopted since the information was from documents in List 3. The category was changed from the previous classification by the use of new information sources. [Evidence Data] (1) Multiple cases of poisoning were reported in humans by oral ingestion of this substance and sodium fluoroacetate, a sodium salt of this substance (CAS RN 62-74-8). As symptoms, there is a description that convulsive seizures, perspiration, and further increases in blood pressure and heart rate occurred following nausea, epigastric pain, and psychiatric symptoms suggestive of auditory hallucinations, etc., resulting in comatose state. Based on these symptoms, the central nervous system and cardiovascular system were considered as the main target organs (ACGIH (7th, 2001)). (2) It was described that in addition to the above symptoms, arrhythmia and ventricular fibrillation appeared in humans by oral ingestion of this substance (GESTIS (Access on September 2019)). (3) It was described that inhalation of aerosols of this substance, even at low concentrations, may cause severe lung damage and neurological or acidosis-related functional disturbances to the respiratory tract due to the development of pulmonary edema and resulting pneumonia (GESTIS (Access on September 2019)). [Reference Data, etc.] (4) The oral lethal doses of this substance in humans were estimated to be within the range of 2-10 mg/kg (HSDB (Access on September 2019)). |

9 | Specific target organ toxicity - Repeated exposure | Category 1 (kidney, reproductive organs (male)) |
Danger |
H372 | P260 P264 P270 P314 P501 |
[Rationale for the Classification] Although there were no data on this substance itself, it was classified based on information on sodium fluoroacetate (CAS RN 62-74-8). Based on (1), it was classified in Category 1 (kidney, reproductive organs (men)). [Evidence Data] (1) In a study in which sodium fluoroacetate was administered by drinking water to rats for 21 days, at or above 6.6 ppm (a converted value equivalent to this substance: 0.83 mg/kg/day (converted guidance value: 0.19 mg/kg/day, within the range of Category 1)), effects on the testes (sperm morphology, sperm cells) and degeneration of the renal tubules were observed (ACGIH (7th, 2001)). [Reference Data, etc.] (2) There was a report that a rabbit exterminator in New Zealand who was exposed during preparation of the bait with fluoroacetic acid over a period of 10 years (details unknown) developed severe and progressive lesions of the renal tubular epithelium and milder hepatic, neurologic and thyroid dysfunctions developed (ACGIH (7th, 2001)). (3) There are reports that in subacute and subchronic tests with rats using sodium fluoroacetate, effects on the testes (degeneration of the seminiferous tubules, hypospermia) and heart were observed at 0.25 mg/kg/day (details unknown) (GESTIS (Access on September 2019)). |
