1 | Acute toxicity (Oral) | Category 4 |
Warning |
H302 | P301+P312 P264 P270 P330 P501 |
Results are reported for single oral administration of 500, 1,000 and 2,000 mg/kg of this substance to 2 rats of each sex. Lethargic tendency and a decrease in body weight were observed in the groups of 500 mg/kg or more. All the rats died in the group of 2,000 mg/kg (JECDB (Access on October 2016)). From the above, the LD50 value is estimated to be more than 500 mg/kg and less than 2,000 mg/kg. Based on this, this substance was classified in Category 4. | FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |

7 | Reproductive toxicity | Category 2 |
Warning |
H361 | P308+P313 P201 P202 P280 P405 P501 |
A combined repeated dose toxicity study with reproduction/developmental toxicity screening test using rats by the oral route (OECD TG 422) was conducted. At the dose (250 mg/kg/day) where the effects on the liver (increase in liver weights, centrilobular hypertrophy of hepatocytes and increased ALT activity etc. ) and the effects on the kidney (increase in kidney weights, dilatation of distal tubule and collecting tubule, and hyperplasia of collecting tubule epithelium) were observed in parental animals, low values in the numbers of live pups at postnatal Days 0 and 4, in the live birth index, and in the body weights of live pups as well as in the viability index on Day 4 were observed in pups (JECDB (Access on November 2016)). Therefore, the substance was classified in Category 2. | FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
8 | Specific target organ toxicity - Single exposure | Category 3 (Narcotic effects) |
Warning |
H336 | P304+P340 P403+P233 P261 P271 P312 P405 P501 |
There were no single-exposure data on this substance in humans. As for experimental animals, it is reported in a single-dose administration study to rats, lethargic tendency and decreased body weights were observed at 500 mg/kg or more of this substance, and all the rats died at 2,000 mg/kg (JECDB (Access on October 2016)). Therefore, this substance was classified in Category 3 (narcotic effects). |

9 | Specific target organ toxicity - Repeated exposure | Classification not possible |
- |
- | - | No information on humans is available. As for experimental animals, in a combined repeated dose toxicity study with reproduction/developmental toxicity screening test (OECD TG 422) using rats administered by gavage, an increase in liver weights was observed at 10 mg/kg/day or more (converted guidance value: 5 mg/kg/day), which is equivalent to Category 1, and centrilobular hypertrophy of hepatocytes was observed at 50 mg/kg/day or more or more(converted guidance value: 23 mg/kg/day), which is equivalent to Category 2 (Access on November 2016)). In addition, an increase in total protein and albumin, an increase in ALT and cholinesterase activity, an increase in kidney weights, dilatation of distal tubules and collecting tubules and hyperplasia of collecting tubule epithelium were observed at 250 mg/kg/day (converted guidance value: 117 mg/kg/day), which is above Category 2 (JECDB (Access on November 2016)). As described above, although the effects on the liver were observed at a dose equivalent to Category 2, they were not severe. Therefore, this substance was classified as "Classification not possible." |
