In a combined repeated dose toxicity study with the reproduction/developmental toxicity screening test by diet administration to rats (OECD TG422, GLP), no adverse effects on sexual function/fertility were observed, but as for the development of offspring, a decreased number of pups delivered per litter, decreased numbers of live pups per litter on postnatal days 1 and 4, increased pup mortality on postnatal day 4, and an increased number of runts were found compared to the control group at the highest dose of 2,000 ppm where general toxicity such as decreased body weights occurred in parent animals (SIDS (2007)). Furthermore, in a developmental toxicity test by oral administration to rats on gestation days 13-15, an increased incidence of malformations such as cleft palate was seen in offspring at or above 229 mg/kg/day where maternal animals showed reduced weight gain, and the increase was significant only at the highest dose of 1,833 mg/kg/day (SIDS (2007)).
