Warning |
H302 | P301+P312 P264 P270 P330 P501 |
The two LD50 values for rats of 1480 mg/kg bw (NTP TR 266 (1988)) and 1480 - 3700 mg/kg bw (IARC vol.53 (1991)) were reported. Based on the value of 1480 mg/kg, the substance was classified as Category 4. | FY2010 | GHS Classification Guidance by the Japanese Government (July, 2010) |
1 | Acute toxicity (Dermal) | Not classified |
- |
- | - | Based on the rabbit LD50 value of > 2500 mg/kg bw (IARC vol.53 (1991)), the substance was classified as "Not classified" in the JIS classification (corresponding to Category 5 or "Not classified" in the UN-GHS classification). | FY2010 | GHS Classification Guidance by the Japanese Government (July, 2010) |

2 | Skin corrosion/irritation | Classification not possible |
- |
- | - | Insufficient data were available. As relevant notes, it was reported that the substance was not irritant to the skin of guinea pigs (HSDB (2002)), however, no details were provided. | FY2010 | GHS Classification Guidance by the Japanese Government (July, 2010) |
3 | Serious eye damage/eye irritation | Classification not possible |

5 | Germ cell mutagenicity | Category 2 |
Warning |
H341 | P308+P313 P201 P202 P280 P405 P501 |
The positive results were reported in the micronucleus test using bone marrow obtained from mice by intraperitoneal administration (in vivo mutagenicity test in somatic cells) (IARC vol.53 (1991)). Besides that, the other positive results were found in the chromosome aberration test using bone marrow obtained from mice by intraperitoneal administration (in vivo mutagenicity test in somatic cells) (IARC vol.53 (1991)). Therefore, the classification was concluded as Category 2. As relevant information, as for in vitro studies, negative results in the Ames test, positive results in the chromosome aberration test using CHO cells and mouse lymphoma assay were reported (NTP DB (Access on Sep. 2010)). | FY2010 | GHS Classification Guidance by the Japanese Government (July, 2010) |
6 | Carcinogenicity | Classification not possible |
- |
- | - | The substance was classified as "Classification not possible" based on the criterion of "Group 3" in the carcinogenicity assessment by the IARC. As relevant information, in the 2-year feeding studies in rats and mice, increases in the incidences of tubular cell adenocarcinoma of the kidney and tubular cell adenomas of the kidney as well as neoplastic nodules or carcinomas of the liver in male rats were reported to be a clear evidence of carcinogenicity. However, it was reported that no evidence of carcinogenicity was found in females in rats and both sexes in mice (NTP TR 266 (1988)). | FY2010 | GHS Classification Guidance by the Japanese Government (July, 2010) |
7 | Reproductive toxicity | Classification not possible |

9 | Specific target organ toxicity - Repeated exposure | Category 2 (blood system) |
Warning |
H373 | P260 P314 P501 |
In the 13-week feeding studies in rats and mice, the effects on the lymphocytes and hematopoietic system were observed in rats treated at 6000 ppm (300 mg/kg/day) or more and in mice treated at 12000 ppm (1800 mg/kg/day) or more, all of which were high doses over the range of guidance values (NTP TR No. 266 (1988)). However, in the 14-month feeding study in male rats administered at 250 - 1000 ppm (12.5 - 50 mg/kg/day), secondary anemia with increased methemoglobin formation and morphological changes of erythrocytes were observed. And hemosiderin deposition was histologically confirmed in the spleen, which was considered as reflecting the response to hemolytic anemia and methemoglobinemia. Pigment deposition was also observed in the kidneys and the liver (HSDB (2002)). Since these changes were seen at the applicable doses to Category 2 of the guidance values, the substance was classified as Category 2 (blood). | FY2010 | GHS Classification Guidance by the Japanese Government (July, 2010) |
