In a reproduction/developmental toxicity screening test with rats dosed by gavage, salivation, cyanosis and delayed parturition were observed in maternal animals at a high dose of 200 mg/kg/day, normal delivery was found only in 1/12 cases, the case of perinatal death which is the total of deaths without delivery or death cases immediately after delivery were observed in 9/12, the remaining 2 cases were also urgently slaughtered on gestational day 25 without delivery. As for humans, it is reported that this substance is a metabolite of the medical drug phenacetin, and it is a causative substance of a blood disorder (methemoglobinemia) on administration of phenacetin (Environmental Risk Assessment for Chemical Substances Vol.8, Tentative Hazard Assessment Sheet (Ministry of the Environment, 2010)).
As for experimental animals, in a 28-day repeated toxicity study with rats dosed by gavage, a decrease in the erythrocyte count, an increase in the reticulocyte count, an increase in the urinary urobilinogen, an increase in the spleen weight, hemosiderin deposition, increased extramedullary hematopoiesis and congestion in the spleen erythroblast hyperplasia in the bone marrow at or above 40 mg/kg/day (converted guidance value: 12 mg/kg/day), and methemoglobinemia, slight hemosiderin deposition and increased extramedullary hematopoiesis in the liver at 160 mg/kg/day (converted guidance value: 50 mg/kg/day) were observed (JECDB (Access on June 2017), Environmental Risk Assessment for Chemical Substances Vol.8, Tentative Hazard Assessment Sheet (Ministry of the Environment, 2010)). In a 4-week repeated inhalation toxicity study with rats (6 hours/day, 5 days/week), an increase in the reticulocyte count, decreases in the hemoglobin concentration, the erythrocyte count and hematocrit value, an increase in the hematopoietic cells in the bone marrow, and hemosiderin deposition in the spleen at or above 86 mg/m3 (converted guidance value: 0.02 mg/L) within the guidance value range (vapour) for Category 1, and decreased body weight gain, an increase in the spleen weight and hemosiderin deposition in the liver, etc. at 883 mg/m3 (converted guidance value: 0.2 mg/L) within the guidance value range (mist) for Category 2 were observed (Environmental Risk Assessment for Chemical Substances Vol.8, Tentative Hazard Assessment Sheet (Ministry of the Environment, 2010)).
From the above, effects on the haemal system and the findings related to them were observed. Therefore, it was classified in Category 1 (haemal system).
