Rationale for the classification Classification year (FY) GHS Classification Guidance for the Japanese Government
1 Acute toxicity (Oral) Not classified
-
-
- - Based on its rat LD50 values of 2650, 2700, and 3195 mg/kg (SIDS (accessed in December 2008)), the substance was rated as "Not classified" using the JIS classification criteria (Category 5 in the United Nations classification). FY2008 GHS Classification Guidance by the Japanese Government (Sep, 2008)
1 Acute toxicity (Dermal) Not classified
-
-
- - Based on its rat LD50 value of 2240 mg/kg (OECD TG401, GLP-compliant) (SIDS (accessed in December 2008)), the substance was rated as "Not classified" using the JIS classification criteria (Category 5 in the United Nations classification). FY2008 GHS Classification Guidance by the Japanese Government (Sep, 2008)
1 Acute toxicity (Inhalation: Gases) Not classified (Not applicable)

Its saturated vapour pressure concentration is 17 mg/m3 (the unit converted). Since its rat LC50 values of 900 mg/m3/4h and > 30000 mg/m3/4h (SIDS (accessed in December 2008)) are higher than its saturated vapour pressure, the substance was classified into Category 3 using criteria for mist, GLP-compliant data and lower LC50 value. In the rabbit tests (OECD TG404, GLP-compliant), the substance was evaluated to be highly irritating and its Primary Dermal Irritation Index value was 8 (which is the maximum score) (SIDS (accessed in December 2008)). In other two tests using rabbits, the substance was concluded to be corrosive (IUCLID (2000)). Based on these results, the substance was classified into Category 1. In other tests using unknown animal species, necrosis was detected 1 minute after the exposure (SIDS (accessed in December 2008), BUA (Report 134, 1993)). Although these results indicate a more refined classification of the substance into Category 1A, further classification was not considered due to lack of information on the species and quantity of animals tested. 3 Serious eye damage/eye irritation Category 1

Danger
H318 P305+P351+P338

P310
Using rabbits, the substance has been documented to be strongly corrosive, and symptoms including hyperemia, and oedema and opacity of the cornea 1 hour after the exposure were detected. These symptoms lasted for 8 days and, additionally, staphyloma were observed (SIDS (accessed in December 2008)). Furthermore, in two other tests using rabbits, the substance was found to be irritating (IUCLID (2000)); and it is rated as Category 1 for skin corrosion and irritation. Thus, the substance was classified into Category 1. FY2008 GHS Classification Guidance by the Japanese Government (Sep, 2008)
4 Respiratory sensitization Classification not possible
-
-
- - No data available. FY2008 GHS Classification Guidance by the Japanese Government (Sep, 2008)
4 Skin sensitization Not classified
-
-
- - In guinea pigs (maximization tests: OECD TG406, GLP-compliant, Buehler tests: non-GLP), the substance was evaluated to be not sensitizing (SIDS (accessed in December 2008)). Thus, it was classified into "Not classified". FY2008 GHS Classification Guidance by the Japanese Government (Sep, 2008)
5 Germ cell mutagenicity Not classified
-
-
- - Since in vivo mutagenicity tests using mouse somatic cells (micronucleus tests) (SIDS (accessed in December 2008)) yielded negative results, the substance was classified into "Not classified". Although positive results were found in micronucleus tests conducted long time ago, the EU-RAR determined that the negative results obtained from above mentioned tests were more reliable. As additional information, in vitro mutagenicity tests (chromosomal aberration tests using Chinese hamsters) yielded positive results (Toxicity Testing Reports of Environmental Chemicals (Chemicals Investigation Promoting Council), accessed in December 2008). FY2008 GHS Classification Guidance by the Japanese Government (Sep, 2008)
6 Carcinogenicity Classification not possible
-
-
- - No data available. FY2008 GHS Classification Guidance by the Japanese Government (Sep, 2008)
7 Reproductive toxicity Classification not possible
-
-
- - In combined repeated oral dose toxicity study with the reproduction/developmental toxicity tests using both female and male rats (20-day forced oral administration during a 2-week period before mating and the gestation period: OECD TG422, GLP-compliant), reproductive or developmental toxicity was not observed (SIDS (accessed in December 2008)). However, since the information on teratogenicity of the offspring was not available, the substance was classified into "Classification not possible". In dermal administration tests using rats of both sexes (1667-2880 mg/kg: OECD TG402), depressions and paresis were observed (SIDS (accessed in December 2008)). Thus, the substance was classified into Category 3 (narcotic effects). FY2008 GHS Classification Guidance by the Japanese Government (Sep, 2008)
9 Specific target organ toxicity - Repeated exposure Classification not possible
-
-
- - In 28-day oral route tests using rats that received doses of 0-1000 mg/kg, epithelial hyperplasia in the mucosa of the urinary bladder was observed in the animal groups of both sexes that received 250 mg/kg (90-day correction value: 38.9 mg/kg/day); hyperplasia of squamous epithelium in the forestomach mucosa was detected in males; and a total serum bilirubin in males and relative weight of the kidney in females increase. However, the change in forestomach is thought to be caused by irritation to the digestive tracts, and the change in the urinary bladder is thought to be caused by a reactive increase by metabolites (Toxicity Testing Reports of Environmental Chemicals (Chemicals Investigation Promoting Council), accessed in December 2008). In other 28-day oral route tests using rats that received doses of 0-500 mg/kg, necrosis in the small intestine mucosa was detected in the animal groups that received 100 mg/kg (90-day correction value: 77.8 mg/kg) (SIDS (accessed in December 2008)). In combined repeated oral dose toxicity study with the reproduction/developmental toxicity tests using rats that received doses of 0-600 mg/kg bw for 40-45 days (OECD TG422, GLP-compliant), the LOEL was 600 mg/kg bw (90-day correction values: 266-300 mg/kg), which is considered to fall under "Not classified" category (SIDS (accessed in December 2008). Based on these results, the substance was classified into "Classification not possible". FY2008 GHS Classification Guidance by the Japanese Government (Sep, 2008)
