In a developmental toxicity test using rats by oral administration on day 6 to 20 of gestation, increased numbers of fetuses with moderate-to-serious skeletal anomalies were observed at the dose where maternal animals showed general toxicity such as decreased body weight gain and increased liver weights. (HSDB (2010)) And in a two-generation reproductive test using rats, a decrease in fetal body weights and a decreased number of the normal offspring judged by skeletal analysis were reported. In a 1-year oral administration test (capsule) in dogs, increases in hemosiderin deposition and extramedullary hematopoiesis in the spleen at 20 mg/kg/day or higher, and anemia with significant decreases in hemoglobin levels, hematocrit, and red blood cell counts, and also elevated platelet counts, brown pigment accumulation in liver reticuloendothelial cells and kidney tubular epithelium, hypercellular sternal bone marrow, and so on at 100 mg/kg/day were observed.
