Oral administration to mice at 136 mg/kg/day, twice a week for 8 weeks caused neuropathy, and revealed adynamic symptom accompanied by slight behavioral changes containing aggression or wariness, and dysfunction of hind limbs. They also showed both atrophy and a significant reduction in the weight of the testes with histopathological degenerations of the epithelial cells of the seminiferous tubules. Thus, toxic effects on the nervous system and testes were noted (EHC 49 (1985), original literature: Hashimoto, K., Sakamoto, J., & Tani, H, (1981) Neurotoxicity of acrylamide and related compounds and their effects on male gonads in mice. Arch. Toxicol., 47: 179 - 189). Here, the converted dose level as that of 90-day study which was calculated from the case of continuous treatment with 136 mg/kg/day for 8 weeks was 83.7 mg/kg/day. Thus, because the exposure amount (twice a week, 8 weeks) in the study was lower than the value, the converted value as that of 90-day study was not exceeded 83.7 mg/kg/day. This means, namely, that the classification could not be applicable to "Not classified" category. The daily dose level in the continuous administration for 8 weeks calculated from the total dose (twice a week for 8 weeks) in this study per capita basis was 38.9 mg/kg/day (converted value as that of 90-day study: 23.9 mg/kg/day). Thus, the exposure dose in this study was considered to be within the range of Category 2 (10 - 100 mg/kg/day) of the guidance values.
