In the acute oral toxicity study in rats (dose levels: 1000, 1400 and 2000 mg/kg), no response to a stimulus within one hour at 1000 mg/kg or higher, followed by prone or lateral position were observed. The degree and duration of such clinical signs were dose dependent. Additionally, increased pericardial fluids and accumulation of pleural fluids were observed at necropsy in dose groups at 1400 and 2000 mg/kg, and besides dark red discoloration of the lungs and liver, whitish opacity or reddening of the mucosa of the glandular stomach ("Toxicity Testing Reports of Environmental Chemicals" (Chemicals Investigation Promoting Council) (Access on Aug. 2010)). From the results described above, the target organ(s) was not specified, but the effects were observed at doses within Category 2 of the guidance values. In the 28-day feeding study in rats (OECD TG 407, GLP-compliant), decreased erythrocyte count, hemoglobin and hematocrit value, extramedullary hematopoiesis and hemosiderosis in the spleen and slightly increased hematopoiesis in the bone marrow in some animals were found in both sexes of the high dose level (1346 mg/kg bw./day, converted dose level as that of 90-day study: 417.26 mg/kg/day). The only treatment-related effect was decreased hematocrit value in females at the mid dose level (273 mg/kg bw./day, converted dose level as that of 90-day study: 84.63 mg/kg/day) (IUCLID (2000)). From the results described above, because the mid dose corresponded to Category 2 of the guidance values, and it was reported that hemolytic anemia was suggested, and that the target organs of toxicity were spleen, bone marrow and erythron (IUCLID (2000)), the substance was classified as Category 2 (hematopoietic system).
