There are no human data. As for experimental animals, in a 90-day diet administration toxicity test using rats, hepatic damage (necrosis of hepatocytes, hypertrophy of parenchyma cells with nuclei of different size, shape, and chromatin content, an increase of hepatocytes having two nuclei, overgrowth of bile duct) was observed at 100 ppm (males: 5.8 to 13.2 mg/kg/day, females: 6.8 to 13.1 mg/kg/day) or higher, and tubulonephrosis in the kidney was found at 800 ppm (males: 52.7 to 98.7 mg/kg/day) or above (ECHA RAC OPINION (2013)). In a 90-day diet administration toxicity test using dogs, fatty degeneration of hepatocytes in the liver and fibrosis of the liver (males) were observed at 60 mg/kg/day or higher (ECHA RAC OPINION (2013)). As above, effects on the liver were found from Category 1, and effects on kidney were observed within a range of Category 2. Therefore, the substance was classified in Category 1 (liver), Category 2 (kidney). |
