There is no information on human reproductive effects. As for experimental animals, in a test in which pregnant rats were dosed by gavage (gestational day 6-20), at 250 mg/kg/day or above where a decrease in body weight gain was observed in maternal animals, lower body weight value was found in the fetuses, and at 500 mg/kg/day or more, in addition to an increase in post-implantation embryo loss, a significant increase in various malformations of the externals, internal organs and skeleton, particularly severe malformations such as cardiovascular malformations, cleft palate, and anal atresia, which were considered not to be related with the maternal toxicity, were observed more frequently than background data (ECHA CLH Report (2011)). In addition, also in the two tests in which pregnant rabbits were dosed by gavage (gestational day 6-28), at 200-220 mg/kg/day where slight suppression of body weight gain was observed in maternal animals, skeletal malformations (malformations of cervical vertebrae/lumbar vertebrae etc.), cardiovascular malformations (ventricular septal defect, subclavian artery defect), or visceral malformations (gallbladder defects, small spleen) were observed (ECHA CLH Report (2011)). Furthermore, also in a test of dermal application to pregnant rabbits (gestational day 6-28), slight suppression of body weight gain and reduced food consumption were observed at 1,000 mg/kg/day in maternal animals, and cardiovascular malformations (subclavian artery defect, ventricular septal defect) occurred at or above 300 mg/kg/day in the fetuses at an incidence exceeding the background data (ECHA CLH Report (2011)).
As described above, in experimental animals, in multiple developmental toxicity tests with pregnant animals, this substance clearly showed teratogenicity in the cardiovascular system, skeleton, internal organs, etc. at doses equal to or lower than the dose where generally slight maternal toxicity was seen. Therefore, it was classified in Category 1B. Besides, the EU classifies this substance as "Repr. 1B" (ECHA CL Inventory (Access on August 2017)).
