In the combined repeated dose toxicity study with the reproduction/developmental toxicity screening test in rats (OECD TG 422, GLP-compliant) (dose levels: 0, 67, 200 and 600 mg/kg/day), effect on reproductive performance of male animals was not observed even at 600 mg/kg group, whereas in females, reduction of the number of corpora lutea of pregnancy was observed at 600 mg/kg, which was considered to cause prolonged gestation length and reduction of implantation number observed at the same dose. For the next generation, numbers of pups born and those born alive were decreased at the 600 mg/kg group ("Toxicity Testing Reports of Environmental Chemicals"(Chemicals Investigation Promoting Council) (Access on Jan. 2011)). In addition, decreased body weight gain was seen in male and female rats at 200 and 600 mg/kg groups ("Toxicity Testing Reports of Environmental Chemicals"(Chemicals Investigation Promoting Council) (Access on Jan. 2011)). The reproductive toxicity was observed at the dose levels, causing general toxicity to parent animals. Therefore, the substance was classified as Category 2. However, since it was reported that these changes were considered as specific phenomenon of male rats with no toxicological significance ("Toxicity Testing Reports of Environmental Chemicals" (Chemicals Investigation Promoting Council) (Access on Jan. 2011)), they were not evidenced to classify. Furthermore, the changes seen at 200 mg/kg/day (converted value as that of 90-day study: approx. 100 mg/kg/day) were suppression in body weight gain in both sexes and increases in weights of both the liver and kidneys in males or females without any other treatment-related changes containing histopathological changes. Thus, "Not classified" was applicable for oral route, but the effects were unknown in the other routes due to no data. Therefore, "Classification not possible" was justified as the classification for specific target organ toxicity (repeated exposure).
