There are no human data. As for experimental animals, in a 28-day gavage administration toxicity test using rats, decreases in hematocrit, hemoglobin content, red blood cell count, cellular infiltration in heart, myocardial degeneration and hypertrophy, hypertrophy of hepatocytes, overgrowth of bile duct, vacuolar degeneration, focal necrosis, and increased mitosis of perilobular hepatocytes in the liver, and hypertrophy of tubular epithelium in renal collecting tube were observed at 0.5 to 12.5 mg/kg/day (converted to a 90-day equivalent value: 0.16 to 3.9 mg/kg/day) within a range of Category 1 (MHLW Japan Existing Chemical Data Base (Access on July 2015)). In a 52-week gavage administration toxicity test using rats, high urine osmolarity, high urine specific gravity, decreases in red blood cell count and hematocrit, increased platelets, prolongation of PT/APTT, increased BUN, increases in A/G, albumin percentage, and ALP, decreases in alpha 2 globulin and beta globulin, decreased alpha 1 globulin, increased absolute/relative liver weight, increased relative kidney weight, centrilobular hypertrophy of hepatocytes in the liver were found at 0.1 to 2.5 mg/kg/day that is within a range of Category 1 (MHLW Japan Existing Chemical Data Base (Access on July 2015)). As above, effects on heart, liver, kidney, and blood were observed within a range of Category 1. Therefore, the substance was classified in Category 1 (heart, liver, kidney, blood system). |
