 As for experimental animals, in an 8-week repeated dose toxicity study using rats given by gavage, a decrease in serum testosterone, an increase in the number of multinucleated giant cells in the testis and an increase in the number of apoptotic cells were observed at 0.03 mg/kg/day (converted guidance value: 0.02 mg/kg/day), which is equivalent to Category 1, and reduction of sperm production and vacuolar spaces in the seminiferous epithelium were observed at 1 mg/kg/day (converted guidance value: 0.5 mg/kg/day) (ATSDR draft (2015)). Also, in a 30-day repeated dose toxicity study using mice given by gavage, effects on the testes (germ cell loss and increased apoptosis) were also observed at 0.045 mg/kg/day (converted guidance value: 0.015 mg/kg/day), which is equivalent to Category 1 (ATSDR draft (2015)). Moreover, in a 30-day repeated dose toxicity study dosed by gavage in rats, effects on the central nervous system (impaired learning and memory, decrease in the density of glutamate receptor in the hippocampus) were reported at 0.1 mg/kg/day (converted guidance value: 0.03 mg/kg/day), which is equivalent to Category 1 (ATSDR draft (2015)).
