- |
- | - | Since no data are available for carcinogenesis evaluation in both humans and experimental animals, and EPA classified this substance as Category D (IRIS Summary (1990)). Therefore, this substance was classified as "Classification not possible" for this hazard class. | FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
7 | Reproductive toxicity | Classification not possible |

No data are available for humans. As for experimental animals, in an 8-week repeated dose toxicity study using rats given by gavage, a decrease in serum testosterone, an increase in the number of multinucleated giant cells in the testis and an increase in the number of apoptotic cells were observed at 0.03 mg/kg/day (converted guidance value: 0.02 mg/kg/day), which is equivalent to Category 1, and reduction of sperm production and vacuolar spaces in the seminiferous epithelium were observed at 1 mg/kg/day (converted guidance value: 0.5 mg/kg/day) (ATSDR draft (2015)). Also, in a 30-day repeated dose toxicity study using mice given by gavage, effects on the testes (germ cell loss and increased apoptosis) were also observed at 0.045 mg/kg/day (converted guidance value: 0.015 mg/kg/day), which is equivalent to Category 1 (ATSDR draft (2015)). Moreover, in a 30-day repeated dose toxicity study dosed by gavage in rats, effects on the central nervous system (impaired learning and memory, decrease in the density of glutamate receptor in the hippocampus) were reported at 0.1 mg/kg/day (converted guidance value: 0.03 mg/kg/day), which is equivalent to Category 1 (ATSDR draft (2015)). Therefore, the substance was classified in Category 1 (genetic organs (men), central nervous system). |

11 | Hazardous to the aquatic environment Short term (Acute) | Category 1 |
Warning |
H400 | P273 P391 P501 |
From 48-hour EC50 = 0.014 mg/L for crustacea (Daphnia magna) (NICNAS, 2001), it was classified in Category 1. It is considered in NICNAS that acute toxicity data for crustacea could be affected by physical effect such as immobilisation which absorption of test substance caused, but it was not evaluated as unreliable data. Therefore, the classification was conducted using the data. Besides, the data on Daphnia magna in NICNAS was from the test of a mixture of tri-, tetra-, penta-, and hexa-forms, but the data was used for the classification because toxicity values were not obtained for a single substance. |

11 | Hazardous to the aquatic environment Long term (Chronic) | Category 1 |
Warning |
H410 | P273 P391 P501 |
If chronic toxicity data are used, then it is classified in Category 1 due to beindg not rapidly degradable (BIOWIN), and ELS NOEC = 0.0089mg/L for fish (Oncorhynchus mykiss) (NICNAS, 2001). If acute toxicity data are used for a trophic level for which chronic toxicity data are not obtained, then it is classified in Category 1 due to being not rapidly degradable (BIOWIN), and 48-hour EC50 = 0.014 mg/L for crustacea (Daphnia magna) (NICNAS, 2001). It was classified in Category 1 from the above results. Besides, the data on Oncorhynchus mykiss and Daphnia magna in NICNAS were from the test of a mixture of tri-, tetra-, penta-, and hexa-forms, but the data was used for the classification because toxicity values were not obtained for a single substance. |
